摘  要:目的探讨青蒿琥酯(Artesunate,Art)是否通过AMPK/mTOR信号通路调控卵巢癌细胞增殖和迁移。方法采用不同浓度青蒿琥酯(12.5,25,50,100mg/L)处理卵巢癌细胞SKOV3,CCK-8实验分析其对肿瘤细胞增殖的影响;划痕实验分析其对肿瘤细胞迁移的影响;Western blot检测各组细胞E-cadherin、Snail及AMPK/mTOR的蛋白表达。结果青蒿琥酯处理的卵巢癌细胞SKOV3活力受到显著抑制,且该效应呈剂量依赖性(P<0.05);青蒿琥酯处理显著抑制卵巢癌细胞SKOV3的迁移能力,上调肿瘤迁移抑制蛋白E-cadherin表达,降低Snail蛋白表达(P<0.05);上调卵巢癌细胞p-AMPK蛋白表达,降低mTOR蛋白表达(P<0.05)。结论青蒿琥酯可通过调控AMPK/mTOR信号通路抑制卵巢癌细胞增殖和迁移。Objective To investigate whether artesunate regulates the proliferation and migration of ovarian cancer cells through AMPK/mTOR signaling pathway.Methods Ovarian cancer cell SKOV3 was treated with different concentrations of artesunate(12.5,25,50,100 mg/L).The effects of artesunate on the proliferation of tumor cells were analyzed by CCK-8 assay.The effects of artesunate on the migration of SKOV3 were analyzed by scratch test.The expressions of E-cadherin,snail and AMPK/mTOR in ovarian cancer cells were detected by Western blot.Results The proliferation of SKOV3 cells treated with artesunate was significantly inhibited in a dose-dependent manner(P<0.05).Artesunate treatment significantly inhibited the migration ability of SKOV3 cells,up-regulated the expression of tumor migration inhibitory protein E-cadherin,and decreased the expression of Snail protein(P<0.05);up-regulated the expression of p-AMPK and down-regulated the expression of mTOR in ovarian cancer cells(P<0.05).Conclusion Artesunate can inhibit the proliferation and migration of SKOV3 cells by regulating AMPK/mTOR signaling pathway.
