摘 要:目的探究PRR15与PI3K/AKT信号通路在三阴性乳腺癌细胞的恶性生物学进程中的作用。方法免疫组织化学方法检测三阴性乳腺癌组织以及癌旁组织中PRR15的表达水平;将三阴性乳腺癌细胞系MDA-MB-468细胞分为pcDNA PRR15组、pcDNA NC组和LY294002组,CCK-8实验检测MDA-MB-468细胞的增殖能力;Transwell实验分析MDA-MB-468细胞的侵袭能力;细胞划痕实验分析MDA-MB-468细胞的迁移能力;蛋白免疫印迹检测PI3K/AKT信号通路以及PRR15蛋白的表达水平。结果三阴性乳腺癌组织中PRR15表达水平降低;pcDNA PRR15和LY294002组MDA-MB-468细胞的生长速度下降、细胞侵袭数量减少、细胞迁移率降低、细胞中PI3K与AKT蛋白表达水平降低。结论PRR15在三阴性乳腺癌组织中低表达,上调PRR15表达后,三阴性乳腺癌MDA-MB-468细胞的增殖、侵袭以及迁移能力降低。Objective To explore the role of PRR15 and PI3K/AKT signaling pathway in the malignant biological process of triple-negative breast cancer cells.Methods The expression level of PRR15 in triple negative breast cancer tissues and adjacent tissues was detected by immunohistochemistry method.Triple-negative breast cancer cell line MDA-MB-468 cells were divided into pcDNA PRR15 group,pcDNA NC group and LY294002 group.The proliferation ability of MDA-MB-468 cells was detected by CCK-8 assay.The invasion ability of MDA-MB-468 cells was analyzed by Transwell assay.The migration ability of MDA-MB-468 cells was analyzed by cell scratch test.The expression levels of PI3K/AKT signaling pathway and PRR15 protein were detected by Western blot.Results The expression of PRR15 was decreased in triple negative breast cancer tissues.In the pcDNA PRR15 and LY294002 groups,the proliferation ability of MDA-MB-468 cells was decreased,the number of cell invasion was decreased,the migration ability was decreased,and the expression level of PI3K and AKT protein was decreased.Conclusion PRR15 is lowly expressed in triple-negative breast cancer tissues.Up-regulation of PRR15 inhibits the proliferation,invasion and migration of MDA-MB-468 cells.
