摘　　要：目的探究JMJD8在肺癌中的表达情况及其对非小细胞肺癌细胞增殖能力影响的潜在机制。方法临床收集87例肺癌组织进行免疫组织化学实验,检测JMJD8的表达情况,并进行JMJD8的表达与临床病理学相关因素之间的统计学分析。MTT实验以及集落形成实验检测JMJD8的表达对细胞的增殖能力的影响。结果JMJD8在肺癌中高表达,与肿瘤的分化程度(P<0.01)和TNM分期(P=0.017)相关。在A549细胞中敲低JMJD8的表达,细胞的增殖能力减弱,并影响增殖相关蛋白的表达。在非小细胞肺癌细胞中过表达JMJD8后,p53及其下游蛋白p21的蛋白表达量下调。结论JMJD8可能作为非小细胞肺癌治疗的新的潜在靶点。Objective To investigate the expression of JMJD8 in lung cancer and its potential mechanism on the proliferation of non-small cell lung cancer cells.Methods 87 tissue samples of lung cancer were collected for immunohistochemical experiments to detect the expression of JMJD8,and the relationship between the expression of JMJD8 and clinicopathological parametors was analyzed.MTT assay and colony assay were used to detect the function of JMJD8 expression on cell proliferation ability.Results JMJD8 was highly expressed in lung cancer,related to the degree of differentiation and TNM stage.After knocking down the expression of JMJD8 in A549 cells,the proliferation ability was weakened,and the expression level of proliferation-related proteins was affected.Over-expressed JMJD8 down-regulated the expression level of p53 and its downstream protein p21 in non-small cell lung cancer cells.Conclusion JMJD8 might be a new potential target for the treatment of non-small cell lung cancer.
