摘 要:目的探究基于Wnt/β-连环蛋白(β-catenin)信号通路的沙参麦冬汤抗小细胞肺癌放射性肺损伤后肺纤维化机制。方法SPF级C57BL/6J小鼠80只,随机分成空白组、照射对照组、照射中药组、抑制剂组,每组20只,建立Lewis肺癌小鼠模型。21d后处死小鼠,称重肺组织,计算肺系数;Masson染色观察各组小鼠肺组织病理学改变;ELISA方法检测肺组织白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、转化生长因子β1(TGF-β1)的表达水平;蛋白免疫印迹法测定小鼠肺组织纤连蛋白(fibronectin)、胶原蛋白Ⅰ型(CollagenⅠ)、α-平滑肌肌动蛋白(α-SMA)、Wnt3a、β-catenin和糖原合成酶激酶-3β(GSK-3β)蛋白表达。结果和空白组比较,照射对照组肺系数增高;肺组织呈现大量胶原沉积,纤维细胞增多;TGF-β1、IL-6、TNF-α、fibronectin、CollagenⅠ、α-SMA水平增高;Wnt/β-catenin信号通路被激活。和照射对照组比较,照射中药组和抑制剂组肺系数降低;肺纤维化程度改善;TGF-β1、IL-6、TNF-α、fibronectin、CollagenⅠ、α-SMA水平降低;Wnt/β-catenin信号通路受到抑制。结论沙参麦冬汤可通过抑制Wnt/β-catenin信号通路活化,对抗小细胞肺癌放射性肺损伤后肺纤维化发挥保护作用。Objective To study the mechanism of Shashen Maidong decoction against lung fibrosis following radiation-induced lung injury in small cell lung cancer based on Wnt/β-catenin signaling pathway.Methods Eighty SPF C57BL/6J male mice were randomly divided into control group,radiation control group,radiation Chinese medicine group and inhibitor group,with 20 mice in each group.Twenty-one days later,the mice were sacrificed,the lung tissue was removed and weighed,and the lung coefficient was calculated.Morphopathological changes in lung tissue were observed by Masson staining.The levels of interleukin-6(IL-6),transforming growth factorβ1(TGF-β1)and tumor necrosis factor-α(TNF-α)in serum were detected by ELISA.The expressions of fibronectin,collagen I,α-smooth muscle actin(α-SMA),Wnt3a,β-catenin and glycogen synthase kinase-3β(GSK-3β)in lung tissues were measured by Western blot.Results Compared with the blank group,the lung coefficient of the irradiation control group was increased.There was a large amount of collagen deposition and fibrocyte increase in lung tissue,and the levels of TGF-β1,IL-6,TNF-α,fibronectin,CollagenⅠandα-SMA were increased.The Wnt/β-catenin signaling pathway was activated.Compared with irradiated control group,the lung coefficient of irradiated Chinese medicine group and inhibitor group was decreased,pulmonary fibrosis was improved,the levels of TGF-β1,IL-6,TNF-α,fibronectin,Collagen I andα-SMA were decreased.The Wnt/β-catenin signaling pathway was inhibited.Conclusion Shashen Maidong decoction can inhibit the activation of Wnt/β-catenin signaling pathway and play a protective role against pulmonary fibrosis after radiation injury of small cell lung cancer.
