摘 要:文章主要研究氧化石墨烯负载精氨酸纳米体系(GO-Arg-SL)对乳腺癌4T1细胞的影响,并对其进行生物安全性的初步评价。选择乳腺癌4T1细胞为研究对象,采用激光共聚焦荧光显微镜和流式细胞术观察GO-Arg-SL对4T1细胞产生NO的影响;采用MTT法和活/死细胞染色法,观察4T1细胞的抑制和杀伤情况,评价GO-Arg-SL对4T1细胞的毒性;采用BALB/c小鼠进行生物安全性实验,观察溶血情况,检测血生化和血常规。结果表明,GO-Arg-SL能使4T1细胞内产生大量的NO;GO-Arg-SL能抑制4T1细胞的生长,加激光照射组的抑制和杀伤能力更强;溶血结果符合规定,血生化和血常规维持在正常范围内。说明GO-Arg-SL光热协同NO气体具有优异的细胞杀伤能力,并且具有良好的生物安全性。To study the effects of graphene oxide supported arginine nanosystem(GO-Arg-SL)on 4T1 cells of breast cancer,and to evaluate its biosafety,the effect of GO-Arg-SL on NO production in 4T1 cells of breast cancer was observed by confocal laser fluores-cence microscopy and flow cytometry.MTT and Calcein AM/PI cell were used to observe the inhibition and killing of 4T1 cells,and the toxicity of GO-Arg-SL to 4T1 cells was evaluated.BALB/c mices were used for biosafety experiment,hemolysis was observed,and blood biochemistry and routine were detected.It was shown GO-Arg-SL can produce a large amount of NO in 4T1 cells.GO-Arg-SL can inhibit the growth of 4T1 cells,and the inhibition and killing ability of laser irradiation group are stronger.The hemolysis results were in accordance with the regulations,and the blood biochemistry and routine were maintained within the normal range.So GO-Arg-SL photothermal synergistic NO gas has excellent cell killing ability,and has good biosafety.
