APP异常表达与口腔鳞状细胞癌生物学行为的关系

摘　　要：探究淀粉样前体蛋白(APP)异常表达与口腔鳞状细胞癌(OSCC)生物学行为的关系。培养人OSCC细胞系WSU-HN13,脂质体转染法建立未感染细胞组、对照质粒组、对照shRNA组、APP过表达组及APP shRNA组,实时定量PCR(qRT-PCR)法检测APP的表达,CCK-8实验检测转染后24、48和72 h细胞增殖活性,细胞划痕实验检测细胞迁移能力,Transwell细胞侵袭实验检测细胞侵袭能力,流式细胞术检测细胞凋亡率。APP过表达组APP mRNA相对表达量明显高于未感染细胞组、对照质粒组、对照shRNA组和APP shRNA组(P<0.05),APP shRNA组APP mRNA相对表达量明显低于未感染细胞组、对照质粒组和对照shRNA组(P<0.05)。转染后24~72 h,APP过表达组细胞增殖活性高于未感染细胞组、对照质粒组、对照shRNA组和APP shRNA组(P<0.05);转染后48~72 h,APP shRNA组细胞增殖活性低于未感染细胞组、对照质粒组和对照shRNA组(P<0.05)。APP过表达组细胞迁移能力、侵袭细胞数高于未感染细胞组、对照质粒组、对照shRNA组和APP shRNA组(P<0.05),APP shRNA组细胞迁移能力、侵袭细胞数低于未感染细胞组、对照质粒组和对照shRNA组(P<0.05)。APP过表达组细胞凋亡率低于未感染细胞组、对照质粒组、对照shRNA组和APP shRNA组(P<0.05),APP shRNA组细胞凋亡率高于未感染细胞组、对照质粒组和对照shRNA组(P<0.05)。研究结果证实APP异常表达可促进OSCC细胞的增殖、迁移、侵袭,抑制细胞凋亡,或可作为OSCC诊疗的新靶点。This study was aimed at investigating the relationship between abnormal expression of amyloid precursor protein(APP)and biological behaviors of oral squamous cell carcinoma(OSCC).The cultured human OSCC WSU-HN13 cells were divided into the uninfected cell group,the control plasmid group,the control shRNA group,the APP over expression group and the APP shRNA group through lipofection transfection.The expression of APP was detected using quantitative real-time polymerase chain reaction(qRT-PCR).The proliferative activity of cells at 24 h,48 h and 72 h after transfection was detected by CCK-8 assay.Cell migration ability was detected by cell wound scratch assay,cell invasion ability was detected by transwell cell invasion assay,and cell apoptosis rate was detected by flow cytometry.The relative expression level of APP mRNA in the APP over expression group was significantly higher than that in the uninfected cell group,the control plasmid group,the control shRNA group,and the APP shRNA group(P<0.05).The relative expression level of APP mRNA in the APP shRNA group was significantly lower than that in the uninfected cell group,the control plasmid group,and the control shRNA group(P<0.05).From 24 h to 72 h after transfection,cell proliferation activity of the APP over expression group was higher than that of the uninfected cell group,the control plasmid group,the control shRNA group and the APP shRNA group(P<0.05).From 48 h to 72 h after transfection,cell proliferation activity of the APP shRNA group was lower than that of the uninfected cell group,the control plasmid group and the control shRNA group(P<0.05).Cell migration ability and invasion cell count in the APP over expression group were higher than those in the uninfected cell group,the control plasmid group,the control shRNA group and the APP shRNA group(P<0.05).Cell migration ability and invasion cell count in the APP shRNA group were lower than those in the uninfected cell group,the control plasmid group and the control shRNA group(P<0.05).The apoptosis rate of
