摘  要:乳腺癌是全球女性最常见的癌症,其发病率和死亡率持续上升,严重威胁着女性的健康。铜是维持人体健康所必需的微量元素,在广泛的生物过程中充当关键的催化辅助因子,其吸收、分布和排出受到严格的调节,从而维持细胞内的铜稳态。铜在乳腺癌治疗中的研究日益兴起,研究表明,铜的动态平衡异常与乳腺癌的发生发展有关。铜死亡是近年来新发现的一种程序性细胞死亡形式,通过铜与三羧酸(tricarboxylic acid,TCA)循环中的脂酰化酶结合从而介导蛋白质聚集和蛋白毒性应激反应并最终导致细胞死亡。本文将基于铜稳态、铜死亡的发生机制作一概述,以探讨铜死亡在乳腺癌发生发展中的作用,以期为乳腺癌临床诊疗提供理论依据与科学指导。Breast cancer is the most common cancer among women worldwide,and its incidence and mortality rates continue to rise,posing a serious threat to women’s health.Copper is a trace element essential for maintaining human health,acting as a key catalytic cofactor in a wide range of biological processes,and its uptake,distribution,and excretion are tightly regulated to maintain intracellular copper homeostasis.Copper is increasingly being researched in the treatment of breast cancer,and studies have shown that an abnormal dynamic balance of copper is associated with the development of breast cancer.Cuproptosis is a newly discovered form of programmed cell death in recent years.It is mediated by copper binding to lipoylases in the tricarboxylic acid(TCA)cycle,which mediates protein aggregation and proteotoxic stress and ultimately leads to cell death.In this paper,we present an overview of copper homeostasis and the mechanism of cuproptosis to explore the role of copper death in the development of breast cancer,with the aim of providing a theoretical basis and scientific guidance for the clinical diagnosis and treatment of breast cancer.
