摘 要:目的:探究2月龄阿尔茨海默症模型小鼠(APP/PS1)初级视皮层(primary visual cortex,V1)神经元在不同视觉刺激模式下的活性变化,解析APP/PS1小鼠早期V1的功能改变。方法:通过免疫荧光染色分析2月龄APP/PS1小鼠初级视皮层内Aβ表达情况,并选取2月龄APP/PS1和野生型(wild type,WT)小鼠,给予其不同条件视觉刺激后,通过c-Fos荧光染色解析V1神经元的活性变化。结果:2月龄APP/PS1小鼠V1内存在Aβ表达但未形成明显淀粉样斑块。在没有外界额外视觉刺激的情况下,2月龄APP/PS1小鼠V1中c-Fos阳性率高于WT小鼠(P<0.01);当给予小鼠外界30分钟的视觉刺激或者视觉刺激后的90分钟,2月龄APP/PS1与WT小鼠V1中c-Fos阳性率均没有明显差异。结论:2月龄APP/PS1小鼠V1神经元在无外界视觉刺激时处于过度活跃状态,其早期视皮层神经元的活动模式可能存在改变。Objective:Investigate the changes of neuronal activity in the primary visual cortex of 2-month-old APP/PS1 mice,an Alzheimer's disease model,under different visual stimulation conditions,and elucidate the early functional alterations in the visual cortex of APP/PS1 mice.Methods:The expression of Aβin the primary visual cortex of 2-month-old APP/PS1 mice was analyzed by immunofluorescence staining,and 2-month-old APP/PS1 and wild type(WT)mice were selected and subjected to different visual stimulation conditions.The changes of neuronal activity in the primary visual cortex were analyzed by performing immunofluorescent staining.Results:Aβis expressed in V1 of 2-month-old APP/PS1 mice,but no obvious amyloid plaques has been found.In the absence of external visual stimulation,the c-Fos positive rate in the V1 of 2-month-old APP/PS1 mice is higher than that in wild-type mice(P<0.01).When given 30 minutes of visual stimulation to mice or 90 minutes after visual stimulation,there is no significant difference in c-Fos positive rate in the V1 of 2-month-old APP/PS1 and WT mice in both cases.Conclusions:The neurons in the V1 of 2-month-old APP/PS1 mice are in a hyperactive state in the absence of external visual stimulation,suggesting altered activity patterns of cortical neurons in the visual cortex of young APP/PS1 mice.
