摘 要:利用慢传输型便秘小鼠模型探讨松二糖的润肠通便作用活性。将昆明小鼠随机分为空白对照(CON,给予生理盐水)组、模型对照(LOP,给予洛哌丁胺)组、阳性对照(POS,给予酚酞和洛哌丁胺)组和松二糖低、中、高剂量干预(LTUR、MTUR和HTUR,给予低、中、高剂量的松二糖和洛哌丁胺)组。连续灌胃7d后,通过排便实验和小肠运动实验评价松二糖的通便效果,并利用聚合酶链式反应、实时荧光定量聚合酶链式反应和酶联免疫吸附方法初步探讨其通便作用机制。结果表明,与LOP组相比,HTUR组[750mg/(kg·d)]的小鼠首粒黑便时间显著缩短了18.04%,粪便含水率增加11.11%,小肠墨汁推进率提高了14.90%;HTUR显著上调便秘小鼠血清中兴奋性神经递质五羟色胺的含量,下调抑制性神经递质血管活性肠肽的含量,同时显著下调诱导型一氧化氮合酶的mRNA表达;此外,与LOP组相比,HTUR组水通道蛋白Aqp3、Aqp4、Aqp8的mRNA的表达水平显著下调,Aqp9的mRNA的表达水平显著上调。综上,松二糖能通过调节神经递质相关因子和水通道蛋白的表达温和有效地缓解洛哌丁胺诱导的慢传输型便秘。The mouse model of slow transit constipation was used to investigate the laxative activity of Tura‐nose.Kunming mice were randomly divided into a blank control(CON,given normal saline)group,a model control(LOP,given loperamide)group,a positive control(POS,given phenolphthalein and loperamide)group,and Turanose intervention(LTUR,MTUR,HTUR,given Turanose and loperamide at low,medium and high doses)groups.After continuous gavage administration for 7 d,the laxative effect of Turanose was evaluated by defecation experiment and small intestinal motility experiment.The laxative mechanism was preliminarily ex‐plored by polymerase chain reaction(PCR),reverse transcription‐polymerase chain reaction(RT‐qPCR),and enzyme‐linked immunosorbent assay.The results showed that:compared with the LOP group,in the HTUR group[750 mg/(kg·d)],the first black stool time significantly shortened by 18.04%,the fecal moisture content increased by 11.11%,and the small intestinal ink propelling rate increased by 14.90%.It was also found that,in the serum of constipated mice,HTUR significantly up‐regulated the content of the excitatory neurotransmit‐ter,serotonin,down‐regulated the content of the inhibitory neurotransmitter,vasoactive intestinal peptide,and down‐regulated the mRNA expression of inducible nitric oxide synthase.In addition,compared with the LOP group,the mRNA expression levels of Aqp3,Aqp4 and Aqp8 were significantly down‐regulated,and the mRNA expression level of Aqp9 was significantly up‐regulated in the HTUR group.In conclusion,Turanose can mildly and effectively relieve loperamide‐induced slow transit constipation by regulating the expression of neurotransmitter‐related factors and aquaporins.
