摘  要:以海藻酸钠为原料,经过氧化法降解、硫酸化修饰得到硫酸化海藻酸钠寡糖。选择氯磺酸‐吡啶体积比、硫酸化试剂‐海藻酸钠寡糖溶液体积比、反应温度和反应时间为单因素,以海藻酸钠寡糖(alginate oligosaccharides,AOS)硫酸化修饰取代度为评价指标,通过响应面试验对氯磺酸‐吡啶法制备硫酸化海藻酸钠寡糖进行优化。确定最佳反应条件为氯磺酸‐吡啶体积比为1∶7,硫酸化试剂与海藻酸钠寡糖溶液体积比为1∶3,反应时间3h,反应温度40℃。在此条件下制备3种不同取代度的硫酸化海藻酸钠寡糖,进行体外抗氧化活性测定。体外抗氧化活性结果表明,硫酸化修饰对AOS的体外抗氧化活性有较大影响,修饰后可以提高AOS的还原力和清除ABTS+自由基、DPPH自由基的能力,且能力随取代度增高而增强。The sulfated alginate oligosaccharides(AOS)were obtained from sodium alginate through oxidative degradation and sulfation modification.The volume ratio of chlorosulfonic acid to pyridine,the volume ratio of sulfating reagent to AOS solution,reaction temperature,and reaction time were selected as single factors.The degree of sulfation substitution of AOS was used as the evaluation index.Response surface methodology was employed to optimize the chlorosulfonic acid‐pyridine method for preparing sulfated AOS.The optimal reaction conditions were determined to be a chlorosulfonic acid‐pyridine volume ratio of 1∶7,a sulfating reagent to AOS solution volume ratio of 1∶3,a reaction time of 3 h,and a reaction temperature of 40℃.Under these conditions,three types of sulfated AOS with different degrees of substitution were prepared and their in vitro antioxidant ac‐tivities were measured.The results of the in vitro antioxidant activity tests indicated that sulfation modification significantly enhanced the antioxidant activity of AOS.The modification improved the reducing power and the ability to scavenge ABTS+and DPPH free radicals,with the effects increasing with higher degrees of substitution.
