奶粉中β-烟酰胺单核苷酸及其类似物的检测

摘　　要：该文建立一种高效液相色谱(high performance liquid chromatography,HPLC)法同时测定奶粉中β-烟酰胺单核糖核苷酸及其类似物的方法。样品经去离子水提取后,添加45 mL乙酸以沉淀蛋白质,经C18固相萃取柱净化后采用Venusil HILIC分析柱分离,β-烟酰胺单核糖核苷酸及其类似物经超纯水、乙腈和0.1%三氟乙酸水溶液梯度洗脱后,采用二极管阵列检测器进行检测。结果表明,该方法检测奶粉中的β-烟酰胺单核苷酸、烟酰胺腺嘌呤二核苷酸、烟酰胺鸟嘌呤二核苷酸、烟酰胺次黄嘌呤二核苷酸、黄素单核苷酸和黄素腺嘌呤二核苷酸在线性范围内的相关系数(R^(2))为0.999 1~0.999 7,方法检出限为0.030 8~0.635 0 mg/kg,定量限为0.102~2.120 mg/kg,回收率为80.4%~101.7%,日内精密度相对标准偏差(relative standard deviation,RSD)(n=6)为1.22%~8.20%。该方法操作简便、准确度高、回收率高和重现性好,适合用于奶粉中烟酰胺单核糖核苷酸及其类似物的检测。A method for determining β-nicotinamide mononucleotide and its analogues in milk powder was established based on high performance liquid chromatography(HPLC).The sample was dissolved in deionized water,and 45 mL acetic acid was added for protein precipitation.The extract was then passed through a C18solid-phase extraction cartridge.Gradient elution was carried out in the Venusil HILIC column with ultrapure water,acetonitrile,and 0.1% trifluoroacetic acid and the compounds were detected by a photo-diode array detector.The results showed that the correlation coefficients(R~2)of β-nicotinamide mononucleotide and its analogues ranged from 0.999 1 to 0.999 7 within the linear range.The method detection limits(MDLs)and method quantification limits(MQLs)were 0.030 8-0.635 0 mg/kg and 0.102-2.120 mg/kg,respectively.The recovery of the compounds varied within the range of 80.4%-101.7%,and the intra-day precision(RSD)(n=6)of the established method was within the range of 1.22%-8.20%.This method is not only simple and cost-effective but also enjoys excellent precision,recovery,and reproducibility.Therefore,it is suitable for determining β-nicotinamide mononucleotide and its analogues in milk powder.
