摘 要:目的分析清胰汤通过调控磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)信号通路对重症急性胰腺炎大鼠的干预效果。方法选取48只健康SPF级SD大鼠,选取8只大鼠分别作为对照组和假手术组,另外32只大鼠建立重症急性胰腺炎模型,其中32只大鼠建模成功,并随机分为模型组、清胰汤组,每组16只。观察4组大鼠病理组织学、淀粉酶、炎性细胞因子、尿素氮、肌酐、超氧化物歧化酶(SOD)、丙二醛(MDA)、PI3K/AKT通路相关mRNA表达量、PI3K/AKT通路。结果与对照组比较,假手术组、模型组、清胰汤组6 h血清淀粉酶、尿素氮、肌酐升高,12 h血清淀粉酶、尿素氮、肌酐升高(P<0.05);与假手术组比较,模型组6 h血清淀粉酶、尿素氮、肌酐升高,12 h血清淀粉酶、尿素氮、肌酐升高,清胰汤组6 h血清淀粉酶、尿素氮、肌酐降低,12 h血清淀粉酶、尿素氮、肌酐降低(P<0.05);与模型组比较,清胰汤组6 h血清淀粉酶、尿素氮、肌酐降低,12 h血清淀粉酶、尿素氮、肌酐降低(P<0.05)。与对照组比较,假手术组、模型组、清胰汤组6 h TNF-α、IL-6、MDA升高,IL-4、SOD降低,12 h TNF-α、IL-6、MDA升高,IL-4、SOD降低(P<0.05);与假手术组比较,模型组6 h TNF-α、IL-6、MDA升高,IL-4、SOD降低,12 h TNF-α、IL-6、MDA升高,IL-4、SOD降低,清胰汤组6 hTNF-α、IL-6、MDA降低,IL-4、SOD升高,12 hTNF-α、IL-6、MDA降低,IL-4、SOD升高(P<0.05);与模型组比较,清胰汤组6 h TNF-α、IL-6、MDA降低,IL-4、SOD升高,12 h TNF-α、IL-6、MDA降低,IL-4、SOD升高(P<0.05)。与对照组比较,假手术组、模型组、清胰汤组PI3K mRNA、AKT mRNA、PI3K、AKT表达量升高(P<0.05);与假手术组比较,模型组、清胰汤组PI3K mRNA、AKT mRNA、PI3K、AKT表达量升高(P<0.05);与模型组比较,清胰汤组PI3K mRNA、AKT mRNA、PI3K、AKT表达量降低(P<0.05)。结论清胰汤可改善重症急性胰腺炎模型大鼠病理组织学,降低炎性反应、尿素氮及肌酐,�Objective To analyze the efficacy of Qingyi Decoction(pancreas clearing decoction)in rats with severe acute pancreatitis through the regulation of phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)signaling.Methods Forty-eight healthy Sprague-Dawley(SD)rats in the specific pathogen-free(SPF)level were selected.Eight rats were randomly selected in the control group(n=4)and sham group(n=4),and 32 rats were selected for severe acute pancreatitis model.Among them,32 rats were successfully modelled,and randomly divided into model group(n=16)and Qingyi Decoction group(n=16).The pathological histology,amylase,inflammatory cytokines,blood urea nitrogen(BUN),creatinine(Cr),superoxide dismutase(SOD),malondialdehyde(MDA),expression levels of proteins in the PI3K/AKT pathway were observed in each group.Results Compared with those of control group,rats in the sham group,model group and Qingyi Decoction group presented significantly higher serum amylase,BUN and Cr at 6h and 12h(P<0.05).Compared with those of sham group,rats in the model group presented significantly higher serum amylase,BUN and Cr at 6h and 12h(P<0.05).Compared with those of sham group,rats in the Qingyi Decoction group presented significantly lower serum amylase,BUN and Cr at 6h and 12h(P<0.05).Compared with those of model group,rats in the Qingyi Decoction group presented significantly lower serum amylase,BUN and Cr at 6h and 12h(P<0.05).Compared with those of control group,rats in the sham group,model group and Qingyi Decoction group presented significantly higher TNF-α,IL-6 and MDA at 6h and 12h,but lower IL-4 and SOD levels at 6h and 12h(P<0.05).Compared with those of sham group,rats in the model group presented significantly higher TNF-α,IL-6 and MDA at 6h and 12h,but lower IL-4 and SOD levels at 6h and 12h(P<0.05).Compared with those of sham group,rats in the Qingyi Decoction group presented significantly lower TNF-α,IL-6 and MDA at 6h and 12h,but higher IL-4 and SOD levels at 6h and 12h(P<0.05).Compared with those of model group,rats in the Q
