摘 要:研究喂食后菲牛蛭水分、蛋白质、氨基酸及抗凝活性的变化规律,为喂食后最佳采收时间提供重要依据。试验选取900条体重约为4 g的菲牛蛭,随机分为5组,每组3个重复,每个重复60条。试验开始当天09:00进行1次喂食,喂食结束后对菲牛蛭禁食,每天14:00分别在每个重复中随机挑选10条进行水分、蛋白质、氨基酸及抗凝活性的测定。结果表明,喂食后第1天菲牛蛭体水分较喂食当天显著降低(P<0.05);喂食后第2天和第4天其体蛋白质含量较喂食当天显著降低4.62%及4.56%(P<0.05);除蛋氨酸、亮氨酸、精氨酸和脯氨酸共4种氨基酸外,其余13种氨基酸含量在喂食当天最高,喂食后第4天最低;随喂食后天数的增加,抗凝活性呈现不断上升的趋势,且喂食后第4天的抗凝活性比喂食当天显著高171.71 U/g(P<0.05)。喂食后不同天数对菲牛蛭水分、蛋白质含量均有显著影响(P<0.05),除蛋氨酸外,其余16种氨基酸含量都差异显著(P<0.05),综合来看,喂食后第3天采收菲牛蛭最佳。To study the changing pattern of moisture,protein,amino acid and anticoagulant activity of Hirudinaria manillensis after feeding,which provides important basis for the optimal harvesting time after feeding.In the experiment,900 Hirudinaria manillensis weighing about 4 g were selected and randomly divided into 5 groups,with 3 replicates in each group and 60 strips in each replicate.One feeding was performed at 9 a.m.on the day of the start of the experiment,and Hirudinaria manillensis were fasted at the end of the feeding,and 10 strips were randomly selected from each replicate at 2 p.m.each day for the determination of moisture,protein,amino acids,and anticoagulant activity,respectively.The results showed that the moisture content of Hirudinaria manillensis on the first day after feeding was significantly lower than that on the day of feeding(P<0.05);the protein content of Hirudinaria manillensis on the second and fourth days after feeding was significantly lower than that on the day of feeding by 4.62%and 4.56%,respectively(P<0.05);and the contents of the other 13 amino acids were the highest on the day of feeding and the lowest on the fourth day after feeding,except for four amino acids,including methionine,leucine,arginine and proline;the anticoagulant activity showed an increasing trend with the increase in the number of days after feeding,and the anticoagulant activity on the fourth day after feeding was significantly higher than that on the day of feeding by 171.71 U/g(P<0.05).Different days after feeding have significant effect on the moisture and protein content of Hirudinaria manillensis(P<0.05),and the contents of the other 16 amino acids are significantly different except methionine(P<0.05),and the Hirudinaria manillensis contains higher anticoagulant activity on the fourth day after feeding.
