摘 要:目的:采用火焰原子吸收分光光度计-AA-7020开发一种测定尿中苯乙烯代谢物浓度的新方法。方法:尿样首先经过2%的硝酸进行酸化处理,随后通过20mL的乙醚进行萃取,萃取液经干燥并重新溶解于5mL的去离子水中后,直接注入火焰原子吸收分光光度计-AA-7020进行检测。在仪器参数设置为波长283.3nm(苯乙烯代谢物如马尿酸的常见检测波长,具体根据实际情况调整)、狭缝宽度0.4nm、燃气流量2.0L/min及助燃气流量1.5L/min的条件下,对尿中苯乙烯代谢物的浓度进行定量分析。评估了该方法的前处理效率、加标回收率(预期范围在90%~110%)、精密度(相对标准偏差预期低于5%)及检出限(预期低于0.05μg/mL)等关键性能指标,并与标准方法进行比较分析。结果:研究中,尿中苯乙烯代谢物浓度与吸光度在0.1~1.0μg/mL范围内呈现良好的线性关系(相关系数不低于0.99),检出限达到0.03μg/mL,加标回收率稳定于95%~105%,相对标准偏差低于3%。精密度方面,将研究建立方法与标准尿检方法进行平行测定对比,测定结果分别为2.5%与2.8%,两种方法在精密度方面无显著性差异(P>0.05)。结论:研究建立的方法在测定尿中苯乙烯代谢物浓度时,展现出了操作简便、灵敏度高、重现性好及适用范围广等优点,为尿中苯乙烯暴露评估提供了一种可靠且高效的检测手段,具有较高的实际应用价值。Objective:To develop a new method for determining the concentration of styrene metabolites in urine using flame atomic absorption spectrophotometer AA-7020.Methods:The urine sample was first acidified with 2%nitric acid,and then extracted with 20 mL of ether.The extract was dried and dissolved in 5 mL of deionized water before being directly injected into the flame atomic absorption spectrophotometer AA-7020 for detection.Quantitative analysis of the concentration of styrene metabolites in urine was conducted under the conditions of instrument parameters set to wavelength 283.3 nm(a common detection wavelength for styrene metabolites such as hippuric acid,adjusted according to actual conditions),slit width of 0.4 nm,gas flow rate of 2.0 L/min,and co gas flow rate of 1.5 L/min.The key performance indicators of this method,including pre-treatment efficiency,spiked recovery rate(expected range of 90%to 110%),precision(expected relative standard deviation below 5%),and detection limit(expected below 0.05μg/mL),were evaluated and compared with the standard method.Results:In the study,the concentration of styrene metabolites in urine showed a good linear relationship with absorbance in the range of 0.1-1.0μg/mL(correlation coefficient not less than 0.99),with a detection limit of 0.03μg/mL.The recovery rate remained stable between 95%and 105%,and the relative standard deviation was less than 3%.Compared with the comparison method,there was no statistically significant difference in the results obtained by the two methods(P>0.05).Conclusion:The method established in this study demonstrates advantages such as simple operation,high sensitivity,good reproducibility,and wide applicability in determining the concentration of styrene metabolites in urine.It provides a reliable and efficient detection method for assessing styrene exposure in urine and has high practical application value.
