膀胱癌中UBE2C、KIF20A蛋白表达与p53基因突变状态的相关性

摘　　要：目的:探究膀胱癌组织中泛素偶联酶2C(UBE2C)、驱动蛋白家族成员20A(KIF20A)蛋白表达情况及其与p53基因突变状态的相关性。方法:选取宿迁市第一人民医院2022年1月~2024年2月住院治疗的120例膀胱癌患者作为研究对象,并均接受p53基因检测。采用免疫组化法检测UBE2C、KIF20A的表达水平,分析UBE2C、KIF20A表达与p53基因突变状态的关系。结果:p53基因突变率与T分期、N分期、肿瘤复发有关联(P<0.05);UBE2C蛋白表达与膀胱癌患者T分期、N分期、肿瘤复发有关联(P<0.05),KIF20A蛋白表达与膀胱癌患者的N分期有关系(P<0.05);膀胱癌患者p53基因突变与UBE2C、KIF20A蛋白表达呈正相关(r=0.181,P=0.048;r=0.311,P=0.001),且UBE2C蛋白表达与KIF20A蛋白表达呈正相关(r=0.562,P<0.001)。结论:UBE2C、KIF20A蛋白的高表达与膀胱癌进展有关,有望成为膀胱癌治疗的靶点和预后评估指标。Objective:To investigate the expression of UBE2C and KIF20A proteins and their correlation with P53 gene mutation in bladder cancer.Methods:120 patients with bladder cancer treated in our hospital were included from January 2022 to February 2024.All patients underwent detection of p53 gene.Immunohistochemistry was used to measure the expression levels of UBE2C and KIF20A.Then the correlation between UBE2C and KIF20A protein expression and mutation status of p53 gene was analyzed.Results:The mutation rate of p53 gene was associated with T and N stage and tumor recurrence(P<0.05).The expression of UBE2C protein was correlated with the T stage,N stage and tumor recurrence in the bladder cancer patients(P<0.05),and KIF20A protein expression was related to N stage(P<0.05).p53 gene mutation in patients with bladder cancer was positively correlated with UBE2C protein expression(r=0.181,P=0.048)and significantly correlated with KIF20A protein expression(r=0.311,P=0.001),and UBE2C protein expression was positively correlated with KIF20A protein expression(r=0.562,P<0.001).Conclusion:The higher expression of UBE2C and KIF20A protein is correlated with progression of bladder cancer.These findings suggest that UBE2C and KIF20A proteins are expected to be a therapeutic target and indicators to estimate the prognosis of patients with this neoplasm.
