摘 要:癌症是世界范围内死亡率较高的严重疾病,主动和被动靶向有助于纳米粒子在肿瘤部位的积累,但是它们的抗癌功效仍受肿瘤部位药物释放不足的限制,这可以通过在肿瘤部位的特异性刺激响应来调节。将刺激响应性材料引入到聚合物纳米载体中,能够预防非特异性药物泄漏,因为它们在特定刺激下释放药物,这赋予药物释放的空间和时间控制性,利用肿瘤微环境调节药物的释放可进一步提高其抗癌功效。近年来,研究者们针对肿瘤微环境设计出了双重和多重响应性智能纳米药物载体。本文主要介绍了基于酶敏感的双响应型聚合物的分类及特性、应用于肿瘤治疗的原理和研究进展。Cancer is a serious disease with a high mortality rate worldwide,and active and passive targeting contributes to the accumulation of nanoparticles at the tumor site,but their anti-cancer efficacy is still limited by insufficient drug release at the tumor site,which can be modulated by specific stimulus responses at the tumor site.The introduction of stimulation-responsive materials into polymer nanocarriers can prevent non-specific drug leakage because they release drugs in response to specific stimuli,which gives spatial and temporal control of drug release,and the use of the tumor micro-environment to regulate drug release can further enhance its anti-cancer efficacy.In recent years,researchers have designed dual and multiple responsive smart nano-drug carriers for the tumor micro-environment.This paper mainly introduces the classification and characteristics of enzyme-sensitive dual-responsive polymers,the principle and research progress of their application in drug targeted therapy.
