摘 要:目的探讨5-脂氧合酶E254K(5-LOX E254K)基因多态性在卵巢癌及健康人群中的表达及意义,血管生长因子(VEGF)在5-LOX E254基因杂合型突变(G/A)和野生型(G/G)卵巢癌血液和组织中的表达及意义。方法采用实时荧光定量PCR法检测101例卵巢癌和101例健康人群血液中5-LOX E254K基因多态性;用酶联免疫吸附法定量检测、免疫组织化学法染色法检测VEGF在卵巢癌患者血浆和组织中的表达;分析两者在卵巢癌中的表达及临床意义。结果(1)5-LOX E254K(G/A)基因杂合突变易患卵巢癌,5-LOX E254K(G/A)基因杂合突变与卵巢癌的发生有着密切的关系,可作为卵巢癌辅助诊断的靶点;(2)5-LOX突变不但与食道、结直肠恶性肿瘤发病相关,还与卵巢恶性肿瘤发病相关;(3)5-LOX E254K基因多态性与淋巴结转移显著相关,5-LOX E254K基因多态性与临床分期、肿瘤大小,发病年龄及病理类型无相关性;5-LOX E254K基因多态性可作为预测肿瘤生物学行为的切入点;(4)VEGF浓度表达与分化程度、卵巢肿瘤标记物CA-125正相关;分化程度与VEGF浓度显著负相关;卵巢肿瘤标记物与VEGF浓度相关,与CA-125显著正相关;与CA-199显著负相关;VEGF浓度与临床分期、肿瘤大小及淋巴转移不存在显著相关;VEGF浓度作为预测卵巢癌发病、分化程度的切入点。(5)5-LOX E254K基因态性在卵巢癌中的表达与VEGF表达无相关性。结论5-LOX E254K基因多态性和VEGF共同促进卵巢癌的发生、发展。Objective To investigate the expression and significance of 5-lipoxygenase E254K(5-LOX E254K)gene polypeptide in ovarian cancer and healthy people,and the expression and significance of vascular endothelial growth factor(VEGF)in 5-LOX E254 gene in blood and tissue of ovarian cancer patients with variant(G/A)and wild type(G/G).Methods The polypeptide of 5-LOX E254K gene was detected by real-time quantitative PCR in 101 cases of ovarian cancer and 101 healthy people.The expression of VEGF in the plasma and tissues of ovarian cancer patients was detected by enzyme-linked immunosorbent assay and immunohistochemical staining.So as to study the expression and clinical significance of the two in ovarian cancer.Results 5-LOX E254K(G/A)gene heterozygous mutation is prone to ovarian cancer,and 5-LOX E254K(G/A)gene heterozygous mutation is closely related to the occurrence of ovarian cancer,and can be used as a target for the diagnosis of ovarian cancer.5-LOX mutations are not only associated with the incidence of esophageal and colorectal malignancies,but also with the incidence of ovarian malignancies.There was significant correlation between 5-LOX E254K gene polymorphism and lymph node metastasis,but no correlation between 5-LOX E254K gene polymorphism and clinical stage,tumor size,age of onset and pathological type.5-LOX E254K gene polymorphism can be used as an entry point to predict the biological behavior of tumors.VEGF concentration and expression were positively correlated with differentiation degree and ovarian tumor marker CA-125.The differentiation degree was negatively correlated with VEGF concentration.Ovarian tumor markers were correlated with VEGF concentration,and significantly positively correlated with CA-125.There was a significant negative correlation with CA-199.There was no significant correlation between VEGF concentration and clinical stage,tumor size and lymphatic metastasis.VEGF concentration can be used as an entry point to predict the incidence and differentiation of ovarian cancer.There was no
