摘  要:目的 探究普萘洛尔对人脐静脉内皮细胞(human umbilical vein endothelial cell,HUVEC)迁移和侵袭的影响,并初步探讨普萘洛尔治疗婴幼儿血管瘤(infantile hemangioma,IH)的可能信号机制。方法 自2022年8月至2024年1月,上海交通大学医学院附属新华医院整形外科以HUVEC细胞系为研究对象,通过划痕实验和Transwell实验观察普萘洛尔及Jagged1等处理前后,对细胞迁移和侵袭能力的影响,通过实时荧光PCR和免疫蛋白印迹实验检测普萘洛尔对Notch信号通路的影响,实验组分为普萘洛尔组、Jagged1+普萘洛尔组,未处理组为空白对照组。结果 普萘洛尔抑制HUVEC在体外的侵袭和迁移,普萘洛尔处理后Notch信号通路成分NICD1、Jagged1和下游效应分子Hes1的表达水平显著下降,Notch1的表达水平显著上调,使用Notch信号通路激活剂Jagged1后部分逆转上述变化。结论 普萘洛尔通过抑制Notch信号通路,显著降低HUVEC的迁移和侵袭能力,这些发现可增强对普萘洛尔治疗IH分子机制的理解。Objective To preliminarily elucidate the potential mechanisms of Propranolol in the treatment of infantile hemangioma(IH) by investigating its effects on the migration and invasion of human umbilical vein endothelial cell(HUVEC) and the possible signaling pathways involved.Methods From August 2022 to January 2024,the HUVEC cell line was used as the research object to observe the effects of Propranolol on cell migration and invasion ability before and after treatment by scratch test and Transwell test.Real-time fluorescent PCR and western blot assay were used to detect the effects of Propranolol on the Notch signaling pathway.The experimental group was divided into the Propranolol treatment group,Propranolol treatment group and Jagged1 treatment group,the untreated group was the blank control group.Results Propranolol significantly inhibited the invasion and migration of HUVEC in vitro.Following Propranolol treatment,the expression levels of Notch signaling components NICD1,Jagged1,and downstream effector Hes1 were markedly decreased,while the expression level of Notch1 was significantly upregulated.These changes were partially reversed by applying the Notch signaling activator Jagged1.Conclusion Propranolol significantly reduces the migration and invasion capabilities of HUVEC by inhibiting the Notch signaling pathway.These findings enhance our understanding of the molecular mechanisms underlying propranolol's therapeutic effects on IH.
