摘 要:木质纤维素作为可再生生物质资源,筛选出高效纤维素降解菌对其利用具有非常重要的意义。从河南农业大学花园中采集土壤样品,通过刚果红染色法、复筛培养基法筛选出一株具有降解纤维素能力的菌株S1-1,通过16s rRNA鉴定,该菌为链霉菌(Streptomyces sp.)。分别测定滤纸酶、内切葡聚糖酶和木聚糖酶的活性,以玉米秸秆为碳源,滤纸酶活性第8天达到最大值0.069 U/mL,内切葡聚糖酶活性第9天达到最大值0.16 U/mL;以玉米芯为碳源,木聚糖酶活性第10天达到最大值26.1 U/mL。玉米秸秆作为碳源时,滤纸酶和内切葡聚糖酶活性较高;玉米芯作为碳源时,木聚糖酶活性较高。本研究获得的菌株S1-1分泌木聚糖酶的活性较高,其分泌的酶系可以作为纤维素酶的辅助酶,从而提高木质纤维素的降解。Wood fiber and cellulose,as renewable biomass resources,have very important significance in selecting efficient cellulose-degrading bacteria for their utilization.A bacterial strain S1-1 with the ability to degrade cellulose was selected from soil samples collected from the garden of Henan Agricultural University through Congo red staining method and secondary screening culture medium method.Through 16s rRNA identification,the bacterium was identified as Streptomyces sp.The activities of filter paper enzyme,endo-glucanase and xylanase were determined,and the strain S1-1 was used as the carbon source.The filter paper enzyme activity reached its maximum value of 0.069 U/mL on the 8th day,the endo-glucanase activity reached its maximum value of 0.16 U/mL on the 9th day.When corn cob was used as the carbon source,the xylanase activity reached its maximum value of 26.1 U/mL on the 10th day.When corn stover was used as the carbon source,the filter paper enzyme and endo-glucanase activities were higher,and when corn cob was used as the carbon source,the xylanase activity was higher.The strain S1-1 produced by this study has high activity of secreted xylanase,and the enzyme system can be used as an auxiliary enzyme to improve the degradation of lignocellulose.
