摘 要:本研究构建了一种电化学检测美洲大蠊多肽1(IPMTAPGI-NH_(2))的新方法。实验以纳米银-二氧化钛-壳聚糖(Ag-TiO_(2)-CS)复合物作为信号放大材料用于修饰电极,通过壳聚糖包埋法将抗体固定在修饰有Ag-TiO_(2)-CS纳米复合物的金电极上,通过多肽和抗体间的特异性反应制备无标记电化学免疫传感器。实验表明,在最优的实验条件下,所设计的电化学免疫传感器线性范围为1~25 ng/mL,检测限可低至0.83 ng/mL。将所建立的新方法应用于美洲大蠊制剂康复新液中该多肽的检测,加标回收率在94.97%~108.1%之间,回收率良好。本研究所构建的电化学免疫传感器简单、快速、灵敏,可用于美洲大蠊制剂中该多肽的测定。In this study,a new method for the electrochemical detection of Periplaneta Americata peptide(IPMTAPGI-NH_(2))was developed.In the experiment,nano-silver-titanium dioxide-chitosan(Ag-TiO_(2)-CS)composite was used as signal amplification material to modify the electrode.Further,the antibody was fixed on the gold electrode modified with Ag-TiO_(2)-CS nanocomposite by chitosan embedding method,and a simple labeling free electrochemical immune sensor was prepared by the specific reaction between the peptide and the antibody.Under the optimal experimental conditions,the linear range of the designed electrochemical immune sensor ranged from 1 to 25 ng/mL,and the detection limit was 0.83 ng/mL.The established method was applied for the detection of the peptide in the Kangfuxin solution,a preparation of Periplaneta Americana.The recovery was between 94.97% and 108.1%.The electrochemical immunosensor constructed in this study is simple,rapid and sensitive,and can be used for the determination of the polypeptide in the preparation of Periplaneta Americana.
