拜城油鸡不同时期脂肪代谢相关基因表达规律的研究

摘  要:为了探讨拜城油鸡胚胎期及出生后1,60日龄脂肪代谢相关基因CCAAT增强子结合蛋白(C/EBPα)、脂肪酸合成酶(FAS)、固醇调节元件结合蛋白1(SREBP1)、过氧化物酶体增殖物激活受体γ(PPARγ)、Spondin3(RSPO3)的表达规律,试验分别采集拜城油鸡和科宝鸡7,9,11,14,17胚龄及出生后1,60日龄胸肌,采用索氏抽提法测定60日龄胸肌中肌内脂肪(IMF)含量,同时检测脂肪代谢相关基因的相对表达量。结果表明:7,11,17胚龄及出生后1日龄拜城油鸡C/EBPα基因相对表达量极显著高于科宝鸡(P<0.01),7~17胚龄及出生后1日龄拜城油鸡FAS基因相对表达量显著或极显著高于科宝鸡(P<0.05或P<0.01),除9胚龄外其他胚龄及出生后1日龄PPARγ基因相对表达量极显著低于科宝鸡(P<0.01),SREBP1和RSPO3基因相对表达量在7~11胚龄时极显著低于科宝鸡(P<0.01)。60日龄时,拜城油鸡FAS和SREBP1基因相对表达量显著低于科宝鸡(P<0.05),RSPO3基因相对表达量极显著低于科宝鸡(P<0.01),C/EBPα和PPARγ基因相对表达量极显著高于科宝鸡(P<0.01)。60日龄拜城油鸡和科宝鸡胸肌IMF含量分别为(3.99±1.33)%和(1.81±0.19)%,拜城油鸡胸肌IMF含量极显著高于科宝鸡(P<0.01)。两个品种鸡60日龄IMF含量分别与C/EBPα、PPARγ基因相对表达量呈中度正相关(P>0.05),与FAS和RSPO3基因相对表达量呈负相关(P>0.05),与SREBP1基因相对表达量呈显著负相关(P<0.05)。说明拜城油鸡和科宝鸡在不同胚龄条件下,C/EBPα、FAS、PPARγ、SREBP1和RSPO3基因表达趋势存在差异。In order to explore the expression pattern of fat metabolism-related genes CCAAT/enhancer-binding proteins(C/EBPα),fatty acid syntheses(FAS),sterol regulatory element binding protein 1(SREBP1),peroxisome proliferator activated receptor(PPAR),Spondin3(RSP03)in the embryonic stage and 60 days of age of Baicheng oil chickens,the breast muscle of Baicheng chickens and Kebao chickens at 7,9,11,14,17 embryonic age,1 day of age and 60 days of age were collected respectively;the content of intramuscular fat in the breast muscle at 60 days of age was measured by Soxhlet extraction method,and the expression of fat metabolism related genes was detected at the same time.The results showed that the expression level of C/EBPαgene in Baicheng oil chickens at the embryonic stage(7,11,17 embryo ages)and 1 day after birth was significantly higher than that of Kebao chickens(P<0.O1);the expression levels of FAS gene in Baicheng oil chickens at the embryonic stage(7-17 embryo ages)and 1 day afer birth was significantly higher than that of Kebao chickens(P<0.05 or P<0.01);the expression level of PPAR gene was significantly lower than that of Kebao chickens at all time points except embryo age 9(P<0.05),and the relative expression levels of SREBP1 and RSPO3 genes were extremely significantly lower than those of Kebao chickens(P<O.O1)from the 7th to the Ilth embryonic ages.At the 60th day of age,the expression levels of FAS and SREBPI genes in Baicheng oil chickens were significantly lower than those of Kebao chickens(P<0.05);the expression level of RSP03 gene at 60 days of age was extremely significantly lower than that of Kebao chicken(P<0.O1),and the expression levels of C/EBPαand PPAR genes were extremely significantly higher than those of Kebao chickens(P<0.01).At 60 days of age,the IMF content in the breast muscle of Baicheng oil chickens and Kebao chickens at 60 days of age was(3.99±1.33)% and(1.81±0.19)%,respectively,and the IMF content in Baicheng oil chickens was significantly higher than that of Kebao chickens(P<0.01).A
