摘 要:目的探究慢性间歇低氧对大鼠胰腺组织、氧化应激及糖代谢的影响。方法将32只成年健康雄性Wistar大鼠以随机数表法分为正常对照组(16只)、慢性间歇低氧组(16只)。正常对照组在常氧下喂养,慢性间歇低氧组循环氮气和氧气8 h/d。6周后建立间歇性低氧模型,第7周第1天处死各组大鼠并留取胰腺组织及血清待用。比较两组丙二醛、超氧化物歧化酶、还原型谷胱甘肽、空腹血糖、空腹胰岛素、糖化血红蛋白、胰岛素抵抗指数、核因子-κB(NF-κB)p65,分析慢性间歇低氧组胰岛素抵抗指数与同组血清氧化应激指标关系。结果6周后经苏木素伊红染色提示慢性间歇低氧组胰腺组织损伤严重。6周后,慢性间歇低氧组丙二醛水平高于正常对照组,超氧化物歧化酶、还原型谷胱甘肽低于正常对照组,差异有统计学意义(P<0.05)。慢性间歇低氧组胰腺组织中NF-κB p65阳性积分高于正常对照组,差异有统计学意义(P<0.05)。慢性间歇低氧组胰岛素抵抗指数高于正常对照组,差异有统计学意义(P<0.05)。经Pearson相关性分析得出,慢性间歇低氧组胰岛素抵抗指数与同组丙二醛呈正相关,与同组超氧化物歧化酶、还原型谷胱甘肽呈负相关(P<0.05)。结论慢性间歇低氧可导致胰腺组织炎症,并且可能通过NF-κB通路引起大鼠氧化应激和胰岛素抵抗。Objective To investigate the effects of chronic intermittent hypoxia on pancreatic tissues,oxidative stress and glucose metabolism in rats.Methods A total of 32 healthy adult male Wistar rats were divided into the normal control group(16 rats)and the chronic intermittent hypoxia group(16 rats)according to randomized digital table method.The normal control group was fed under normoxia conditions,and the chronic intermittent hypoxia group was fed under nitrogen and oxygen cycling for 8 h/d.The intermittent hypoxia model was established after 6 weeks.The rats in each group were executed on Day 1 of Week 7.Their pancreatic tissues and serums were obtained and retained for use.The malondialdehyde,superoxide dismutase,reduced glutathione,fasting glucose,fasting insulin,glycated hemoglobin,insulin resistance index,nuclear factor-κB(NF-κB)p65 were compared between the two groups.The relationship between insulin resistance index in the chronic intermittent hypoxia group and serum oxidative stress index in the same group was analyzed.Results Severe pancreatic tissue damage was indicated by hematoxylin eosin staining in the chronic intermittent hypoxia group after 6 weeks.The malondialdehyde level was higher,and the superoxide dismutase and reduced glutathione were lower in the chronic intermittent hypoxia group than those in the normal control group after 6 weeks,with statistically significant differences(P<0.05).The NF-κB p65 positive integration in pancreatic tissues was higher in the chronic intermittent hypoxia group than that in the normal control group,with a statistically significant difference(P<0.05).The insulin resistance index was higher in the chronic intermittent hypoxia group than that in the normal control group,with a statistically significant difference(P<0.05).Pearson correlation analysis showed that the insulin resistance index in the chronic intermittent hypoxia group was positively correlated with malondialdehyde in the same group,and negatively correlated with superoxide dismutase and reduced gluta
