摘  要:铜是人体必需的微量金属元素之一,广泛参与了体内多种生命活动过程,包括氧化还原反应、酶促反应、能量代谢及营养物质代谢等.铜稳态的维持对于细胞正常发挥功能至关重要,细胞内铜缺乏或过量都会引起细胞功能异常,进而引发疾病.在肿瘤发生发展过程中,铜通过调控细胞代谢及信号转导过程影响细胞增殖和血管生成,进而参与肿瘤的侵袭和转移.对铜稳态维持的基本过程进行综述,探讨铜稳态异常参与肿瘤关键信号通路调控的作用机制,并介绍3种已应用于肿瘤治疗的靶向铜稳态相关药物,分析它们的作用机制及临床应用潜力.Copper is one of the essential trace metals in the human body,playing a key role in various biological processes,including redox reactions,enzymatic reactions,energy metabolism,and nutrient metabolism.The maintenance of copper homeostasis is crucial for the normal functioning of cells,as both copper deficiency and excess can lead to cellular dysfunction and disease.During tumor development,copper can influence cell proliferation and angiogenesis by regulating cellular metabolism and signal transduction pathways,thus contributing to tumor invasion and metastasis.This review aims to summarize the key processes involved in maintaining copper homeostasis,explore the mechanisms by which copper homeostasis disruption contributes to the regulation of key signaling pathways in human cancers,and introduce three copper homeostasis-targeting drugs currently applied in cancer treatment,analyzing their mechanisms and clinical potential.
