高效液相色谱法测定白芷酒中香豆素类成分的方法研究

摘  要:为了建立能够准确测定白芷大曲酒中含有的3种香豆素类化合物含量的方法,对高效液相色谱法检测香豆素类化合物的色谱条件进行了优化研究。方法:将酒样进行超声处理后,经过微孔滤膜过滤,以水(A)、乙腈(B)为流动相进行梯度洗脱,流速为1 mL/min,检测波长和柱温分别为254 nm、30℃,外标法定量。结果:在该检测方法下3种香豆素类化合物的线性关系良好,其中佛手柑内酯R^(2)=0.9986,定量限浓度为0.143μg/mL,平均回收率为103.6%;欧前胡素R^(2)=0.9983,定量限浓度为0.039μg/mL,平均回收率为100.3%;异欧前胡素R^(2)=0.9919,其对应的定量限浓度以及平均回收率分别为0.127μg/mL、99.5%。结论:通过此检测方法开展测定工作,能够快速制备样品,保证测定结果的精准性,同时重复性以及稳定性较好,因此,此方法测定白芷大曲酒中3种香豆素类化合物十分适用。In order to establish a method capable of accurately determining the contents of three coumarins contained in Angelica dahurica daiquiri,the chromatographic conditions for the detection of coumarins by high performance liquid chromatography(HPLC)were optimized.Methods:The wine samples were ultrasonicated and then filtered through a microporous filter membrane,and the gradient elution was carried out with water(A)and acetonitrile(B)as the mobile phases at the flow rate of 1 mL/min,and the detection wavelength and column temperature were 254 nm and 30 C,respectively,and the external standard method was used for quantification.Results:The linearity of the three coumarin analogs was good under this assay,including bergamot lactone R^(2)=0.9986,the limit of quantification(LOQ)concentration of 0.143μg/mL,and the average recovery of 103.6%;euoprostenol R^(2)=0.9983,the limit of quantification(LOQ)concentration of 0.039μg/mL,and the average recovery of 100.3%;isoepistephamenol R^(2)=0.9919,with a limit of quantification(LOQ)of 0.127μg/mL and an average recovery of 99.5%,respectively.Conclusion:This method is suitable for the determination of three coumarins in Angelica dahurica daiquiri because it can prepare the sample quickly and ensure the accuracy of the results with good reproducibility and stability.
