大蒜素对脑局灶缺血大鼠血浆中血小板颗粒膜蛋白-140和血管性血友病因子水平的影响

摘　　要：目的观察大蒜素对大鼠大脑中动脉血栓模型血浆GMP-140和vWF水平的影响。方法采用三氯化铁致大脑中动脉血栓(MCAT)模型,观察药物的作用;并测定造模后动物血小板的聚集性、血浆中GMP-140与vWF水平及药物对其的影响。结果不同剂量的大蒜素均能明显改善MCAT模型大鼠的神经症状,缩小大鼠脑梗塞范围;另外,大蒜素体内给药对ADP和胶原诱导的血小板聚集有明显的抑制作用,可以显著降低模型大鼠血浆中GMP-140与vWF的水平。结论大蒜素可能通过抑制血小板活化而降低局灶性缺血大鼠的脑损伤。Objective:To study the effects of garlicin on mi ddle cerebral artery thr om-bosis(MCAT)rats and its influence on the plasma c ontent of GMP -140an d vWF.Methods:Middle cerebral Thrombosis(MCAT)model was made with FeCl 3 .The platelet aggregation rate,the plasma contents of GMP -140and vWF of the animal were measured,and the neurologic deficit(ND)and the infarct siz e(IS)of the model were observed.Results:Garlicin of high and low dose(5, 10mg /kg)attenuated the ND of rats subjected t o MCAT.IS of the low dose gr oup and high dose group with MCAT decreases by 53.93%and 60.70%,res pectively.Garlicin could also inhibit platelet aggregation induced by col and A DP,a nd reduce the plasma contents of GMP -140and vWF of the MCAT rats.Con clusion:Garlicin could reduce the brain injury in rats with focal cerebral isch emia,and the possible me ch-anism is to inhibit the activation of platelet.
