摘  要:目的:作者分别采用酶消化法和组织块法培养人牙周膜细胞,并对这两种方法进行比较。方法:倒置相差显微镜连续观察体外培养的人牙周膜细胞原代和传代生长情况,从形态学、免疫组化及细胞增殖方面对比两种方法培养细胞的异同。结果:两种方法培养的细胞形态大致相同,来源一致,增殖和生长曲线大体一致。组织块法培养的细胞同源性好,但初代培养时间长;酶消化法培养的细胞一次可获得的细胞量大,但容易污染,且初代培养的细胞混有较多的上皮细胞,需多次传代才能去除。结论:组织块法培养HPLFs适用于细胞纯度要求较高的实验,而酶消化法培养HPLFs 适用于短期内获得大量细胞的实验。Objective: To compare explant techniques and monolayer culture in culturing human periodontal ligament fibroblasts. Methods: Cell cultures were performed from human periodontal ligament by means of explant techniques and monolayer culture. Cells were characterized on the basis of morphological, biochemical, and proliferative parameters. Result: HPLFs cultured in two different methods were similar in maphology and proliferation. Cells cultured in explant techniques needed a long period of time to confluent in the primary culture; Cells obtained from monolayer culture were mixed with epithelial cells and must be purified by several processes of passive culture. Conclusion: Explant techniques used to culture HPLFs can help us obtain comparatively pure cells, but the primary culture needs a long period of time. Cells obtained from monolayer culture are more than from explant techniques in a short period of time, but we have to purify cells by successive passage culture.
