内皮素-1诱导培养的人肺动脉平滑肌细胞I、Ⅲ型胶原蛋白及其mRNA表达

摘　　要：为研究内皮素 1(ET 1)对培养的人肺动脉平滑肌细胞 (HPASMC )I、Ⅲ型胶原合成的影响 ,应用贴块法培养人胎儿肺动脉平滑肌细胞 ,应用免疫细胞化学染色、原位杂交方法检测I、Ⅲ型胶原蛋白及其mRNA的表达。结果显示 ,加入含 10 - 8mol LET 1的培养液培养 4 8h和 72h后 ,HPASMCI、Ⅲ型前胶原mRNA及其蛋白表达明显增强。提示ET 1能够诱导培养的人肺动脉平滑肌细胞I、Ⅲ型胶原合成增加 ,其调控机制之一是在转录水平上增强了I。To investigate the effect of endothelin 1 on Type I,Ⅲ collagen synthesis in cultured human pulmonary artery smooth muscle cells (HPASMCs). Human fetal pulmonary artery smooth muscle cells were cultured using explant method. The changes of type I,Ⅲ collagens and relevant mRNA were examined by immunocytochemistry and in situ hybridization . The results showed that type I,Ⅲ collagens and relevant mRNA contents increased significantly after treatment with 10 -8 mol/L ET 1 for 72 and 48 hours. In conclusion, ET 1 can stimulate Type I,Ⅲ collagen synthesis in cultured HPASMCs by promoting the transcription of relevant mRNA.
