摘  要:目的:探讨MMP-1,MMP-2,MT1-MMP和MT2-MMP在NASH肝纤维化中的相互作用及可能机制.方法:采用原位分子杂交方法检测40例NASH患者的肝穿刺标本中MMP-1,MMP-2,MT1-MMP和MT2-MMPmRNA的表达.结果:MMP-1,MMP-2,MT1-MMP和MT2-MMPmRNA主要表达在纤维间隔和汇管区的间质细胞(肝星状细胞等),且MMP-2和MT1-MMP的表达细胞有重叠.NASH组MMP-2,MT1-MMP,MT2-MMPmRNA的表达均低于ASH组(P=0.03,P=0.007,P=0.001)结论:MMP-1,MMP-2,MT1-MMP和MT2-MMP在NASH肝纤维化的发生和发展中可能起重要作用;MMP-2和MT1-MMP在基质降解过程中可能有协同作用;肝星状细胞为肝组织内MMP-1,MMP-2,MT1-MMP和MT2-MMP的主要产生细胞.AIM:To explore the relationship among the expression ofMMP-1,MMP-2,MT1-MMP and MT2-MMP in the liver tissueswith NASH and the mechanism of hepatic fibrosis.METHODS:Expressions of MMP-1,MMP-2,MT1-MMP andMT2-MMP mRNA in 40 cases of liver tissues with nonalco-holic steatohepatitis (NASH) were detected with in situhybridization.RESULTS:MMP-1,MMP-2,MT1-MMP and MT2-MMP mRNAwere expressed mainly in mesenchymal cells (such as he-patic stellate cell, HSC) in the fibrous septa and portal tract.Meanwhile,some cells were positively stained both the MMP-2and the MT1-MMP mRNA. The expression of MMP-2, MT1-MMPand MT2-MMP in the liver tissues with NASH was lower thanthat with ASH, respectively (P =0.03,P =0.007,P =0.001).CONCLUSION:Expression of MMP-1, MMP-2, MT1-MMP andMT2-MMP may play an important role in the development ofliver fibrosis of NASH; MMP-2 may be coordinate with MT1-MMP in the degradation of extracellular matrix (ECM); HSCseems to be the major source cells of MMP-1, MMP-2, MT1-MMPand MT2-MMP in NASH.
