摘  要:目的 探讨高温对肿瘤细胞周期的影响。方法 将肝癌腹水型瘤株HepA细胞分为 3 7℃、40℃、42℃和 44℃四个温度组。每一组分别在 1h、2h、4h和 8h四个时间点取材 ,经PI染色在流式细胞仪上检测四种温度条件下 ,四个时间段时HepA细胞在各细胞周期中的细胞数。结果  3 7℃时 ,细胞处于活跃的增殖状态 ,绝大部分细胞处于S及G2 /M期。 40℃时 ,细胞增殖更趋活跃 ,G1期细胞减少。 42℃时 ,细胞增殖受到明显抑制 ,G1期细胞增加 ,G2 /M期细胞减少。 44℃时 ,12h ,细胞增殖受到进一步的抑制 ,G1期细胞明显增加 ,G2 /M期细胞进一步减少 ;48h ,高温对细胞增殖的影响反而减弱 ,与 12h时相比 ,G1期细胞明显减少 ,G2 /M期的细胞有了明显的增加 ,甚至多于 42℃时。结论 温和性高温 (4 2 44℃ )对HepA细胞的增殖活动有明显的抑制作用 。Objective To study the effects of hyperthermia on the cell cycle of tumor cells. Methods The HepA cells were divided into four groups, and placed respectively in 37℃, 40℃, 42℃ and 44℃. Each group was heated for 1, 2, 4 and 8 hours, and tested by the flow cytometer. Results At 37℃, the cells proliferated actively and most cells were in S phase and G2/M phase. At 40℃, the cells proliferated more actively and the number of cells in G1 phase was decreased. At 42℃, the proliferation of the cells was inhibited. The number of cells in G1 phase was increased and that in G2/M phase decreased. At 44℃ for 1 2 hours, the inhibition effect of hyperthermia on cell proliferation was enhanced. The number of cells in G1 phase was increased and that in G2/M phase decreased further. At 44℃ for 4 8 hours, the inhibition effect of hyperthermia on cell proliferation was weakened. The number of cells in G1 phase were decreased and that in G2/M phase increased markedly than at 44℃ for 1 2 hours. Conclusion The moderate hyperthermia(42 44℃) has a remarkable inhibition effect on proliferation of HepA cells and keeps them arrested in the G1 phase.
