摘 要:目的研究Pokemon及鼠双染色体2(MDM2)在食管鳞状细胞癌(ESCC)组织中的表达及临床意义。方法采用免疫组化法检测93例ESCC组织、30例癌旁正常黏膜组织(对照)中Pokemon及鼠双微染色体2(murine double minute 2,MDM2)蛋白的表达水平,分析Pokemon及MDM2蛋白表达与临床病理各参数之间的关系。结果 Pokemon在对照组织和ESCC中的阳性表达率分别为6.67%(2/30)和64.52%(60/93)(P<0.01);MDM2在对照组织和ESCC中的阳性表达率分别为20%(6/30)和58.06%(54/93)(P<0.01)。Pokemon蛋白表达水平与ESCC淋巴结的转移和临床病理分期呈正相关(P<0.05),而与患者的年龄、性别、组织学分级和浸润深度等无相关(P>0.05)。MDM2蛋白的表达与ESCC的组织学分级、淋巴结转移和临床分期呈正相关(P<0.05),而与患者的年龄、性别、和浸润深度无相关性(P>0.05)。Pokemon和MDM2在ESCC组织中呈正相关(r=0.508)。结论 Pokemon和MDM2的激活促进了ESCC的发生发展、浸润及转移。联合检测两基因的表达对深入了解食管癌的发展及转移机制具有重要意义。Objective To study the expression and clinical significance of Pokemon and MDM2 in esophageal squamous cell carcinoma(ESCC).Methods Immunohistochemical staining was used to detect the expression of Pokemon and MDM2 proteins in 93 cases of ESCC tissues and 30 cases of adjacent normal tissues.The relationship between Pokemon and MDM2 and the pathological characteristics of ESCC was analyzed.Results The positive rate of Pokemon in control and ESCC tissues was 6.67%(2/30)and 64.52%(60/93),respectively(P<0.01).The rate of MDM2 in control and ESCC tissues was 20%(6/30)and 58.06%(54/93),respectively(P<0.01).The expression of Pokemon was correlated positively with lymph node metastasis TNM stage,but not significantly correlaed with patients' age,gender,diameter and differentiation of ESCC tissues(P>0.05).The expression of MDM2 protein was correlated positively with lymph node metastasis,differentiation and TNM stage(P<0.05),but not significantly correlaed with patients' age,gender and diameter of ESCC tissues(P>0.05).Furthermore,the expression of Pokemon was correlated positively with that of MDM2(r=0.508,P<0.05).Conclusion Pokemon and MDM2 may play an important role in the occurrence,development,invasion and metastasis of ESCC.Codetecting Pokemon and MDM2 proteins is very importment to understand deeply the mechanism of progression and metastsis of ESCC.
