摘 要:目的利用四氯二苯并二恶英(tetrachlorodibenzo-p-dioxin,TCDD)建立稳定高效的昆明小鼠腭裂模型。方法采用不同剂量的TCDD作用于不同妊娠天数的昆明小鼠,观察胎鼠腭裂的发生情况以及药物的毒性影响,确定诱导形成腭裂的最佳条件。结果孕12.5天的昆明小鼠给予一次性灌胃40μg/kg TCDD,可诱导胎鼠腭裂发生率为98.04%。显微镜下观察:13.5天时,实验组及对照组侧腭突位于舌两侧;14.5天时,对照组侧腭突增大并上抬,呈对向生长,头部开始融合;实验组侧腭突增大,未出现上抬以及融合。15.5和16.5天时,对照组侧腭突完全融合,实验组侧腭突上抬至舌上方,但体积较小,未出现融合,形成腭裂。16.5天时,胎鼠的肝、肺无明显组织学结构异常。结论昆明小鼠于孕12.5天一次性灌胃40μg/kg TCDD可建立稳定高效的小鼠腭裂动物模型;TCDD主要通过延迟侧腭突上抬诱导昆明小鼠腭裂发生。Objective To establish a stable and efficient animal model of cleft palate induced by tetrachlorodibenzo-p-dioxin( TCDD) in Kunming mice. Methods To determine the optimum conditions of cleft palate formation,we observed the cleft palate occurrence and toxic effects in fetal mice with different doses of TCDD at different gestation days( GD). Results The incidence rate of cleft palate was 98. 04% with 40 ug / kg TCDD by gavage at GD12. 5. By microscopic observations,lateral palatine processes were located on the lateral sides of the tongue in both groups at GD13. 5. The enlarged lateral palatine processes were elevated above the tongue and began to fuse with each other from the head end in the control group at GD14. 5. In the experimental group,the lateral palatine processes were enlarged,but not elevated above the tongue,so no fusion was found. The lateral palatine processes had completely fused with each other in the control group at GD15. 5 and GD16. 5. In the experimental group,the lateral palatine processes were elevated above the tongue,but they were smaller and no fusion was found. The cleft palate had formed. No obvious abnormality was found in liver and lung in fetal mice at GD16. 5. Conclusion Stable and efficient animal model of cleft palate was induced with 40 ug / kg TCDD by gavage in Kunming mice at GD 12. 5. TCDD induced cleft palate information mainly by delaying the elevation of lateral palatine processes in Kunming mice.
