摘  要:目的 研究梁金菇多糖 (ABPS)对人白血病细胞K5 62凋亡的影响及其机制。方法 应用荧光显微镜观察凋亡细胞形态 ;应用流式细胞仪检测ABPS对K 5 62细胞周期的改变及凋亡的影响 ;应用Westernblot检测bcl -6及增殖细胞核抗原 (PC NA )的表达情况。结果 荧光显微镜下观察到K 5 62细胞核碎裂、核溶解和凋亡小体形成等典型的凋亡形态学变化。DNA凝胶电泳观察到”阶梯状”条带。用药 5天后 ,G1期细胞数明显增多 ,占 5 4.5 % ,42 .0 %的细胞发生凋亡。bcl -6表达水平升高 ,PCNA表达水平下降。结论 ABPS能诱导K 5 62细胞发生凋亡 ,其机制可能与PCNA表达水平下降、bcl-6表达水平升高有关。Objective To study the effect of Agaricus blazei polysaccharide(ABPS) on apoptosis in K562 cells of leukemia.Methods Morphology of apoptotic cells was studied by fluorescent microscope;cell cycle phases and number of apoptotic cell were analyzed by flow cytometry;bcl 6 and proliferation cell nuclear antigen(PCNA) were checked by Western blot.Results Typical apoptotic cells was observed by fluorescent microscopy and typical DNA ladder appeared on agar gel;Number of K562 cell increased significantly in G 1 phase,and the number of apoptotic cell was 42.0% at the 5th day;bcl 6 was increased and PCNA was decreased in K562 cells at the 7th day.Conclusion ABPS can induce K562 cells occur typical apoptosis,the mechanism might be related to cell cycle arrest and the change of PCNA and bcl 6.
