土贝母皂甙对单疱病毒性角膜炎作用的实验研究

摘　　要：目的　通过动物试验评价土贝母皂甙 (tubeim oside,Tu)治疗单疱病毒性角膜炎(herpes simplex keratitis,HSK)的效果及滴眼刺激性。方法　在地鼠肾 BHK- 2 1传代细胞上进行单纯疱疹病毒 - 1型 (herpes sim plex virus- 1,HSV- 1) SM4 4毒株复苏 ,制作兔 HSK模型。采用裂隙灯显微镜、扫描电镜和透射电镜观察眼部病变的动态变化评价 Tu疗效 ,同时进行实验评价 Tu滴眼刺激性。结果　 Tu联合应用贝复舒或无环鸟苷可以提高 Tu疗效 ,Tu提前用药类似于联合应用贝复舒。 0 .1g· L- 1 Tu与生理盐水眼刺激分值的统计学结果无显著性差异。结论　 Tu治疗实验性 HSK有一定的疗效 ,对 HSK具有预防作用。联合应用无环鸟苷不能增强无环鸟苷抗病毒能力 ,联合应用贝复舒可以增强 Tu治疗 HSK的疗效。 0 .1g· L- 1Objective To evaluate the therapeutic effect of tubeimoside (Tu) on eyes of rabbit herpes simplex keratitis (HSK) models in depth and its irritational effect on normal rabbit eyes.Methods Herpes simplex virus 1(SM44 strain) was propagated and quantitated on monolayers of BHK 21 secondary cell cultures so that rabbit models of HSK could be made after viral inoculation with scarification. All eyes were examined by slit lamp microscopy and selective corneal samples were observed under scanning and transmission electron microscope. Meanwhile Tu was topically administered to assess its irritational effect on normal rabbit eyes. Results Response to treatment of Tu combined with basic fibroblast growth factor(bFGF), or combined with acyclovir (ACV), was better than that attained with Tu alone. Effect obtained with Tu prematurely beginning medication after one hour post inoculation was similar to that of Tu combined with bFGF. By contrast with those of normal saline (NS), statistical results of Tu showed no significant difference of ocular irritational score.Conclusion Tu is able to cure experimental HSK to a certain degree, also can prevent its occurrence and development. Tu combined with ACV can not enhance ACV antiviral potency, but combined with bFGF, Tu can get better cural effect. 0 1g·L -1 Tu given topically brings about no irritational effect.
