牛磺酸对血小板活化因子灌注离体大鼠肾损害的保护作用

摘  要:目的 :探讨血小板活化因子 (PAF)对离体大鼠肾损害的作用机制和牛磺酸的保护作用 .方法 :实验大鼠均建成离体肾脏灌流模型 ,用K -H灌流液作对照组 ,余两组在K -H液中分别加PAF ,PAF及牛磺酸灌流 ,检测灌流液中的蛋白含量、组织钙、丙二醛 (MDA) .结果 :PAF组漏出蛋白量、组织钙、MDA明显高于对照组 (P <0 0 1) ;牛磺酸组漏出蛋白量明显低于PAF组 (P <0 0 1) ,组织钙、MDA低于PAF组 (P <0 0 5 ) .结论 :PAF是通过其受体影响细胞膜钙通道开放 ,钙内流增加 ,导致钙超载 ,PAF促进细胞膜脂质过氧化损伤肾脏 ;牛磺酸通过调节细胞内钙稳态作用 。Objective: To observe the effect of platelet activating factor (PAF) and protective role of Taurine on the kidney of rat in vitro. Methods: The experimental animal models were divided into 3 groups by kidney infusing in vitro using PAF, PAF+Taurine, the K-H solution infusing was used as control. The amounts of protein, calcium and malondialdehyde (MDA) in the tissue were measured and used as the observation indexes. The results indicated that the amounts of protein, calcium and malondialdehyde in the group of PAF were markedly higher than the control(P<0 01). In the group of PAF+Taurine, the amounts of protein were much lower than the group of PAF(P<0 01),the amounts of calcium and malondialdehyde were also much lower(P<0 05). Conclusions: Our study suggests that the mechanism of PAFs effect on the kidney of rat is to affect the acceptor on the membrane, which opens the calcium channel and increases the calcium inflow and the lipid peroxidation that might injure the kidney of rat; Taurine could protect the kidney of rat by adjusting the intracellular calcium and inhibiting the lipid peroxidation.
