摘  要:我们用甲基紫精为唯一碳源、氮源的选择培养基,从长期使用百草枯的农田土样中筛选出一株可降解甲基紫精的细菌菌株XT12,利用分光光度法检测了菌株XT12的生长曲线和培养液中甲基紫精的含量。一方面,我们发现菌株XT12可以降解甲基紫精,并且其对甲基紫精的降解率随甲基紫精初始浓度的增大而减小:当甲基紫精的初始浓度分别为5μg/m L、10μg/m L、20μg/m L时,菌株XT12对甲基紫精的降解率分别为40.58%、29.00%、22.15%;另一方面,我们发现甲基紫精可以缩短菌株XT12的对数生长期,使其提前达到稳定期。通过形态学特征、理化性质、16S r RNA序列及进化树分析,我们发现菌株XT12属于高地芽孢杆菌(Bacillus altitudinis),将其命名为Bacillus altitudinis lbh XT12。综上所述,高地芽孢杆菌XT12有望用来改善被百草枯污染的水土环境。We screened the strains that can degrade methyl viologen with methyl viologen as the sole carbon source and nitrogen source from the long-term use of paraquat of farmland soil sample. We have obtained a biodegradable methyl viologen bacterial XT12. We detected the growth curve of XT12 and the content of methyl viologen in the broth by spectrophotometry. On one hand, We found the strain XT12 can degrade methyl viologen,and its degradation rate to methyl viologen decreases with the increase of initial concentration of methyl viologen.When the initial concentration of methyl viologen are 5μg/m L, 10 μg/m L, 20μg/m L, the degradation rate of methyl viologen to the strain XT12 is respectively 40.58%, 29.00%, 22.15%. On the other hand, We found that methyl viologen can shorten the logarithmic growth phase of the strain XT12 and make the strain XT12 to achieve a stable period in advance. We identified the strain XT12 by morphological characteristics, physical and chemical properties and 16 S r RNA sequence. We found that the strain XT12 belongs to the Bacillus altitudinis. We named the strain XT12 Bacillus altitudinis lbh XT12. In summary, the strain XT12 is expected to be used to improve the contaminated soil and water by paraquat.
