鬼臼毒素衍生物GP-14对HL-60细胞凋亡及端粒酶活性的影响

摘　　要：目的　观察自旋标记鬼臼毒素衍生物 4 β -N -〔4″- (2″,2″ ,6″ ,6″ -四甲基 - 1″ ,2″ ,5″,6″ -四氢吡啶 - 1″ -氮氧自由基 ) -酰胺基〕 - 4′ -去甲基表鬼臼毒素 (GP - 14 )对人白血病细胞系HL - 6 0的作用及对端粒酶活性的影响。方法　采用MTT法、电镜、琼脂糖凝胶电泳和流式细胞仪检测GP - 14对HL - 6 0细胞的作用 ;TRAP -ELISA法测定端粒酶活性。结果　GP - 14可明显抑制HL - 6 0细胞增殖。在一定剂量和时间范围内 ,GP - 14主要诱导HL - 6 0细胞凋亡 ,5 0 μmol/L处理 4 8小时凋亡率为 4 3 8% ;并可使端粒酶活性下降。 结论　GP - 14能特异地诱导HL - 6 0细胞凋亡 ,抑制细胞端粒酶活性 ,对端粒酶活性的抑制作用比同等浓度的VP - 16强。为进一步开发其临床价值提供了依据。ObjectiveTo investigate the antitumor activities and apoptosis mechanisms of 4β-N-〔4″-(2″,2″,6″,6″-tetramethyl-1″,2″,5″,6″-tetrahydrogenpyridine-1″-nitroxyl)-amido〕-4′-demethylepipodophyllotoxin(GP-14) on HL-60 cells.MethodsMTT colorimetric assay,electron microscope,agar gel electrophoresis and flow cytometry were used to detect the effects of GP-14 on HL-60 cells.The telomerase activity was detected by TRAP-ELISA assay.ResultsThe growth of HL-60 cells were suppressed apparently by GP-14.In some extent of dosage and time,GP-14 induced the apoptosis of HL-60 cells.The apoptosis rate was 43.8% incubated by 5.0μmol/L GP-14 for 48 hours.GP-14 resulted in the inhibition of telomeraseactivity.ConclusionGP-14 could induce the apoptosis of HL-60,inhibit telomeraseactivity specifically.It's inhibitation was stronger than that of VP-16 with same concentration,which provided basis for further study for clinical use.
