摘  要:目的:研究大肠癌胃泌素(gastrinGAS)、生长抑素(somatostatinSS)mRNA及其蛋白表达之间的相关性.方法:取手术切除的大肠癌新鲜标本50例,采用逆转录巢式PCR(RT-PCR)检测GAS、SSmRNA,同时用免疫组化方法检测相应标本GAS、SS蛋白表达.结果:大肠癌GAS、SSmRNA及其蛋白表达呈显著正相关,(r=0.9304和0.9405,P<0.01);中、低分化及中晚期大肠癌GAS表达高,(χ2=7.07,P<0.01和χ2=4.12,P<0.05),尤其以黏液腺癌、印戒细胞癌最为显著;中、低分化大肠癌生长抑素表达低,(χ2=8.59,P<0.05).结论:大肠癌自分泌GAS、SS调节大肠癌生长;中、低分化大肠癌,尤其黏液腺癌和印戒细胞癌应作为大肠癌内分泌治疗的主要适应证.AIM:To investigate the correlation between the mRNAs andproteins expression of gastrin (GAS) and somatostain (SS)in colorectal carcinoma.METHODS:Tumor tissues in 50 cases of colorectal carci-noma were studied by using the reverse transcriptase-poly-merase chain reaction (RT-PCR) and immunohistochemicaltechniques.RESULTS:There was a significant positive correlation be-tween the expression of mRNAs and proteins of GAS andSS in colorectal carcinoma (r =0.9304 and 0.9405, P <0.01 );The positive rates of gastrin expression of moderately andpoorly differentiated cancers were significantly higher thanthat of well differentiated cancer (χ2 =7.07, P<0.01 and χ2=4.12, P <0.05 ). Expression of gastrin in mucinous adeno-carcinoma and signet-ring cell carcinoma was significantlyhigher than those in papillary and tubular adenocarcinomas;The positive rates of somatostatin expression in moderatelyand poorly differentiated cancers were significantly lowerthan that in well differentiated one.CONCLUSION:Colorectal carcinoma secretes GAS and SSto regulate its growth. The moderately and poorlydifferentiated, mucinous adenocarcinomas and signet-ringcell carcinoma of the colorectum may be the candidates forendocrine therapy.
