摘  要:目的 探讨重型肝炎患者的血浆模型在生物人工肝研究中的应用价值。方法 将血浆置换手术中无菌收集的重型肝炎患者的血浆作为重型肝炎模型 ,在体外研究了猪肝细胞型生物人工肝对重型肝炎血浆生化指标及药物转化代谢的影响。结果 重型肝炎患者的被置换血浆经过猪肝细胞型生物人工肝的处理后 ,血浆中的总胆红素、谷氨酰转肽酶、胆碱酯酶、血糖、胆固醇及糖化血清蛋白的水平出现下降 ,总蛋白、白蛋白、甘油三酯水平增加 ,加入血浆中的氨茶碱浓度迅速下降。结论 该血浆模型能反映出生物人工肝对糖、蛋白质、脂肪等物质代谢产生的影响及对药物的转化代谢活性。重型肝炎患者的被置换血浆作为重型肝炎模型 ,对于生物人工肝的疗效研究具有重要的应用价值。Objective To study the role of plasma exchanged from the patient with severe hepatitis in the research of bioartificial liver as a model of severe hepatitis.Methods The plasma was aseptically collected from the patient with severe hepatitis during the treatment of plasma exchange operation.The effect of bioartificial liver with porcine hepatocytes on the biochemical changes and biotransformation of aminophylline was investigated by the circulating of the plasma in vitro.Results The levels of total serum bilirubin,γ glutamyl transpeptidase,cholinesterase,glucose,cholesterol,serum glycoprotein in the plasma from the patient with severe hepatitis were lowerd and the levels of total protein,albumin,triglycerides were elevated after the treatment of bioartificial liver by circulating in vitro.The content of aminophylline was decreased rapidly after the treatment.Conclusion This plasma model of severe hepatitis is sensitive to the treatment of bioartificial liver on the metabolism of sugar,fat and protein and drug biotransformation.It is valuable to the study of the bioartificial liver on severe hepatitis.
