摘 要:目的 观察电刺激小脑顶核(FN)后感觉皮层PKCγ和PKCδ表达的变化,以探讨电刺激FN预置性神经保护作用产生的机制。方法 建立大鼠电刺激小脑顶核模型,分别在电刺激后即刻(Oh)、1d、3d、7d、10d取感觉皮层区脑组织,冰冻切片,分别进行PKCγ和PKCδ免疫组化染色。图像分析测定其平均光密度值。并设立假刺激组及小脑齿状核(DN)刺激组作为对照。结果 一侧电刺激FN后即刻,对侧感觉皮层PKCγ和PKCδ表达无明显变化(P>O.05),而1d后PKCγ和PKCδ表达显著增高(P<0.01);3d后其表达有所降低,7d后降至基础水平。同侧感觉皮层在电刺激FN后1d,PKCγ和PKCδ表达也有显著增高(P<O.05),同样持续至第3天,7d后其表达亦降至基础水平,但其增高程度明显低于刺激对侧(P<O.O5)。假刺激组及DN刺激后1d、PKCγ和PKCδ表达无明显改变。结论 电刺激FN诱导的预置性神经保护作用的产生与PKCγ和PKCδ有关。Objective To investigate the mechanism of the preconditioned neuroprotection of cerebellar fasti-gial nucleus (FN) electrical stimulation,and the effect of electrical stimulation at FN on expression of PKCγ and PKCδ in somatosensory cortex of rats. Methods The FN stimulation models of rats were established. Brain tissue containing somatosensory cortex was removed at Oh, Id,3d,7d and lOd after FN stimulation respectively and 20μm coronal sections were obtained using a sliding cryo-microtome. Two sections of each rat were processed for im-munocytochemical analysis of PKCγ and PKCS and the intensity of immunostaining was quantified by a computer-assisted image analysis system. The pseudo-stimulated rats and the cerebral dentate nucleus(DN) stimulated rats were used as control groups. Results Immediatly after FN stimulation, the expressions of PKCγ and PKCδin contralateral somatosensory cortex were not changed. But both elevated much significantly at Id and persisted for 3 days. In 7 days they decreased to basal level of pseudo-stimulated control group. The expressions of PKCγand PKCδ in ipsilateral somatosensory cortex were also elevated at Id and lasted for 3 days after FN stimulation, but the elevation was significantly lighter than that in contralateral ones. There was no change of PKCγ and PKCδexpression in pseudo-stimulated and DN-stimulated groups. Conclusion It is suggested that the production of preconditioned neuroprotection of FN stimulation may be related to the PKCγand PKCδ expression.
