摘 要:目的 研究99Tcm 氮 二 (N 乙基 N 乙氧基二硫代氨基甲酸盐 ) [N(NOEt) 2 ]在人宫颈癌Hela细胞中的摄取动力学。方法 用放射性核素示踪法研究人宫颈癌Hela细胞在 37℃和 2 2℃时对99TcmN(NOEt) 2 和99Tcm 甲氧基异丁基异腈 (MIBI)的摄取动力学。结果 Hela细胞摄取99TcmN(NOEt) 2的峰值为 4 6 .15 % ,99Tcm MIBI为 12 .6 0 % (P <0 .0 1) ;5min时Hela细胞摄取99TcmN(NOEt) 2 为峰值的6 5 % ,99Tcm MIBI为 5 0 % (P <0 .0 5 ) ;6 0min时99TcmN(NOEt) 2 在Hela细胞中的滞留率为 6 6 0 2 % ,99Tcm MIBI为 5 6 .6 7% (P <0 .0 5 ) ;Hela细胞对99TcmN(NOEt) 2 的摄取不依赖温度 ,99Tcm MIBI摄取则呈温度依赖性。结论 99TcmN(NOEt) 2 可能较99Tcm MIBI更适用于肿瘤显像 ,有潜在的临床应用价值。ObjectiveTo investigate the cellular kinetics of 99Tcm-bis(N-ethoxy-N-ethyl dithiocarbamato)-nitrido N(NOEt) 2 in human cervical carcinoma Hela cells and to compare it with the cellular kinetics of 99Tcm-methoxyisobutylisonitrile (MIBI) in similar conditions; to define the possible clinical value of 99TcmN(NOEt) 2 in tumor imaging. MethodsAccording to the principle of radionuclide tracer technique, 99TcmN(NOEt) 2 or 99Tcm-MIBI were incubated with Hela cells at 37 ℃ or at 22 ℃, at several time points after incubation, the uptake and washout characteristics of radiotracers in Hela cells were investigated and compared. ResultsThe maximum uptake of 99TcmN(NOEt) 2 in Hela cells was 46.15%, that of 99Tcm-MIBI was 12.60%(P<0.01). The uptake of 99TcmN(NOEt) 2 after 5 min incubation in Hela cells was 65% of total uptake, uptake of 99Tcm-MIBI was 50% of total uptake( P<0.05); 1 h after removal of the unbound tracer, 66.02% of the total uptake of 99TcmN(NOEt) 2 was retained in the Hela cells, 56.67% of 99Tcm-MIBI was retained (P<0.05); not like 99Tcm-MIBI, the cellular kinetics of 99TcmN(NOEt) 2 was not temperature-dependent (the cellular kinetics was similar at 37 ℃ and at 22 ℃, P>0.05). ConclusionIn vitro data suggest that 99TcmN(NOEt) 2 may be a tracer better than 99Tcm-MIBI for tumor imaging; 99TcmN(NOEt) 2 has potential application in clinical use.
