大鼠角膜移植术后早期角膜基质细胞凋亡的初步研究

摘　　要：目的　研究角膜移植术后早期角膜移植物基质细胞凋亡发生的规律。方法　建立大鼠穿透性角膜移植动物模型 ,以同基因角膜移植和同种异体角膜移植为实验组 ,并以正常大鼠做正常对照。术后一周取材 ,应用光镜、电镜及细胞凋亡原位末端标记 (TUNEL法 )分别检测植片伤口附近 (<0 3mm)及植片中央区 (>0 5mm)角膜基质细胞凋亡情况。结果　对照组和实验组角膜中央区基质细胞凋亡数无明显差异 ;实验组角膜植片伤口附近基质细胞凋亡明显增加 ,两实验组相比无明显差异。结论　角膜移植术后一周基质细胞凋亡主要发生在切口附近 ,可能与伤口愈合有关。Objective To study apoptosis of keratocytes in corneal grafts during the early postoperative period. Methods Orthotopical corneal transplantation model was developed. Two groups were allograft (Wistar to SD rats) and autograft (Wistar to Wistar rats) as experimental groups, and the normal rats (Wistar rats) as control. A week after operation the corneal samples were removed and fixed. The apoptosis in keratocyte was studied by light and electron microscopes and TUNEL assay. Results The TUNEL-positive cells of keratocytes in the control group were 0.20±0.10(near the margin) and 0.17±0.12(in the central), which was consistent with the allograft and the autograft groups in the central donor corneas. But around the wound (<0.3 mm) the TUNEL-positive cells of the allograft and the autograft groups were significantly higher compared with the control group(P<0.01), respectively. Conclusion Apoptosis is frequent in the corneal grafts around the wound a week after transplantation, which may be related to the wound healing during the earlypostoperative period.
