摘 要:目的 研究柴胡皂苷D对胃癌细胞糖酵解代谢及其肿瘤生物学特性的影响。方法 采用不同浓度的柴胡皂苷D(0.0,0.5,1.0,2.0μmol/L)处理胃癌细胞AGS及MGC803;CCK8实验检测药物处理后胃癌细胞的增殖活力、划痕实验检测胃癌细胞迁徙能力、Transwell实验检测胃癌细胞侵袭能力;采用乳酸试剂盒检测胃癌细胞糖酵解代谢终产物乳酸的代谢水平;通过GEPIA在线分析TCGA和GTEx数据库中原癌基因(YAP1、c-Myc)在胃癌中表达;Western印迹检测胃癌细胞YAP1、c-Myc蛋白表达。结果 柴胡皂苷D显著抑制胃癌细胞AGS及MGC803增殖、迁移及侵袭(均P<0.05);显著抑制胃癌细胞糖酵解代谢终产物乳酸的生成水平(P<0.05)。生物信息数据库统计显示,YAP1、c-Myc在胃癌组织中表达显著高于胃正常组织(均P<0.05),柴胡皂苷D显著抑制AGS及MGC803中YAP1、c-Myc蛋白表达(均P<0.05)。结论 柴胡皂苷D能够抑制胃癌细胞部分肿瘤生物学表型,其机制可能与其抑制了胃癌细胞糖酵解代谢及癌基因YAP1、c-Myc蛋白表达相关。Objective To study the effect of Saikosaponin D on glycolytic metabolism and oncobiology characteristics in gastric cancer cells.Methods Different concentrations of Saikosaponin D(0.0,0.5,1.0,2.0μmol/L)were used to deal with AGS and MGC803 gastric cancer cells;CCK8 experiment was used to detect proliferation activity of gastric cancer cells,scratches experiment was used to test migration ability of gastric cancer cells,Transwell method was used to detect invasion ability of gastric cancer cells;the metabolic level of lactic acid,which was the end product of glycolysis metabolism was detected by lactic acid kit in gastric cancer cells;expressions of protooncogene(YAP1,c-Myc)in TCGA and GTEx databases were analyzed by GEPIA online website in gastric cancer;Western blot was used to detect YAP1 and c-Myc protein expressions in gastric cancer.Results Proliferation,migration,invasion of AGS and MGC803 gastric cancer cells were significantly inhibited by Saikosaponin D(all P<0.05).The production level of lactic acid,which was the end product of glycolysis metabolism was significantly inhibited in gastric cancer cells(P<0.05).Biological infor-mation database statistics showed that expressions of c-Myc and YAP1 were significantly higher in gastric cancer tissue than those in normal gastric tissue(all P<0.05).YAP1 and c-Myc protein expression levels of AGS and MGC803 cells were significantly inhibited by Saikosaponin D(all P<0.05).Conclusions Saikosaponin D could inhibit partial tumor biological phenotypes of gastric cancer cells,the mechanism may be that Saikosaponin D could inhibit glycolytic metabolism and oncogene YAP1 and c-Myc protein expres-sions of gastric cancer cells.
