KLF15基因多态性与大动脉粥样硬化型缺血性脑卒中的相关性分析

摘　　要：目的探讨Krüppel样因子15(KLF15)基因rs9838915位点的多态性与大动脉粥样硬化(LAA)型缺血性脑卒中的相关性。方法采用病例-对照研究方法,选取2021年6月—2022年6月在新疆医科大学第二附属医院就诊的新发LAA型缺血性脑卒中患者105例为病例组,同期在该院体检健康者103名为对照组。收集两组的临床资料,并采用飞行质谱分析技术检测KLF15基因rs9838915位点的基因型。计算基因型和等位基因频率分布,并与对照组进行比较。结果病例组的C反应蛋白、白细胞介素-6、中性粒细胞与淋巴细胞比值单核细胞和高密度脂蛋白胆固醇比值、血小板与淋巴细胞比值、D-二聚体水平均高于对照组,而高密度脂蛋白胆固醇水平低于对照组(P<0.05)。病例组和对照组KLF15基因rs9838915位点的基因型分别为GG、AA、GA,频率分布病例组分别为8.65%、33.65%和57.69%,对照组分别为7.00%、38.00%和55.00%。等位基因G、A在病例组的频率分布分别为74.52%和25.48%,在对照组分别为74.00%和26.00%,两组KLF15基因rs9838915位点基因型和等位基因频率分布比较,差异无统计学意义(P>0.05)。在共显性模型、显性模型、隐性模型、超显性模型及加性模型分析中发现,KLF15基因rs9838915位点与LAA型缺血性脑卒中无相关性(P>0.05)。结论KLF15基因rs9838915位点多态性与LAA型缺血性脑卒中无相关性,未来需要更多的研究进一步探讨这一多态性的潜在机制和作用。Objective To analyze the correlation between polymorphism at rs9838915 site of Krüppel-like factor 15(KLF15)gene and large artery atherosclerotic(LAA)ischemic stroke.Methods A method of case-control study was conducted to select 105 patients with newly diagnosed LAA ischemic stroke who were treated in the Second Affiliated Hospital of Xinjiang Medical University from June 2021 to June 2022 as the case group,while 103 healthy people who underwent physical examinations in this hospital during the same period were selected as the control group.The clinical data was collected in the two groups,and the genotype of KLF15 gene rs9838915 site was detected by flight mass spectrometry.The genotype and allele frequency distribution were calculated and compared with the those in control group.Results The levels of C-reactive protein,interleukin-6,neutrophil to lymphocyte ratio,monocyte to high-density lipoprotein cholesterol ratio,platelet to lymphocyte ratio and D-dimer level in the case group were significantly higher than those in the control group,while the level of high-density lipoprotein cholesterol was significantly lower than that in the control group(P<0.05).The genotypes of KLF15 gene rs9838915 site in the case and control groups were GG,AA and GA,respectively,and the frequencies distributions of the case group were 8.65%,33.65%,and 57.69%,while those in the control group were 7.00%,38.00%,and 55.00%,respectively.The frequency distributions of alleles G and A in the case group were 74.52%and 25.48%,while those in the control group were 74.00%and 26.00%,respectively.There were no significant differences in the genotype and allele frequency distribution of KLF15 gene rs9838915 between the two groups(P>0.05).There was no significant correlation between KLF15 gene rs9838915 site and LAA ischemic stroke by analyses of co-dominant model,dominant model,recessive model,super-dominant model and additive model(P>0.05).Conclusions The rs9838915 site polymorphism of the KLF15 gene is not related to LAA ischemic stroke in the
