摘 要:目的:研究沙参四味汤诱导人肺癌NCI-H460细胞的凋亡及其机制。方法:采用免疫组化方法检测不同浓度沙参四味汤作用后的肺癌NCI-H460细胞GADD45a的表达,测定GADD45a在细胞内的范围及数量;应用流式细胞术(FCM)技术检测细胞凋亡改变;应用荧光定量PCR方法检测Caspase-3和Caspase-7基因表达变化。结果:沙参四味汤组GADD45a显著升高,不同浓度沙参四味汤组与对照组之间均有统计学差异。FCM观察Annexin-V/PI双染结果显示,沙参四味汤组细胞发生的凋亡率较对照组增多(P<0.05)。荧光定量PCR检测发现,与对照组比较,沙参四味汤组NCI-H460细胞Caspase-3和Caspase-7基因表达降低(P<0.05)。结论:沙参四味汤对人肺癌NCI-H460细胞具有潜在的体外增殖抑制作用,沙参四味汤诱导肺癌细胞中GADD45a、Caspase-3和Caspase-7产生,促使人肺癌NCI-H460细胞凋亡,因此推断沙参四味汤能抑制肺癌H460细胞生长。Objective:To investigate the apoptosis-inducing effects of Shashensiwei decoction on human lung cancer NCI-H460 cells and to explore the underlying mechanism of apoptosis.Methods:The expression of GADD45a in lung cancer NCI-H460 cells treated with different concentrations of Shashensiwei decoction was detected using immunohistochemistry.The range and quantity of GADD45a in the cells were measured.Flow cytometry(FCM)was used to assess changes in cell apoptosis.Fluorescent quantitative PCR was employed to detect changes in the expression of Caspase-3 and Caspase-7 genes.Results:The expression of GADD45a was significantly increased in the Shashensiwei decoction group,with statistical differences observed between the Shashensiwei decoction groups of different concentrations and the control group.FCM analysis of Annexin-V/PI double staining results showed that the apoptosis rate of cells in the Shashensiwei decoction group was higher compared to the control group(P<0.05).Fluorescent quantitative PCR revealed that the expression of Caspase-3 and Caspase-7 genes in NCI-H460 cells was decreased in the Shashensiwei decoction group compared to the control group(P<0.05).Conclusion:Shashensiwei decoction exhibits potential in vitro inhibitory effects on the proliferation of human lung cancer NCI-H460 cells.It induces the production of GADD45a,Caspase-3,and Caspase-7 in lung cancer cells,promoting apoptosis in NCI-H460 cells.Therefore,it is inferred that Shashensiwei decoction can inhibit the growth of lung cancer H460 cells.
