摘  要:结直肠癌是人类常见的恶性肿瘤。N6-甲基腺嘌呤(N6-methyladenosine,m6A)是真核生物信使RNA(Messenger RNA,mRNA)中主要的内部表观遗传修饰,具有动态可逆的特点。在转录后mRNA的的剪接、翻译和降解调控中起关键决定作用。m^(6)A甲基转移酶在肿瘤的发生发展中发挥促癌或抑癌的作用,并且具有成为恶性肿瘤治疗靶点的潜力。甲基转移酶样3(Methyltransferase-like 3,METTL3)是最重要最主要的m6A甲基转移酶,多项研究表明其通过多种机制发挥促进结直肠癌的作用。本文对METTL3在结直肠癌发生发展中的作用和相关机制进行综述,旨在为结直肠癌进一步寻找生物标记物和治疗靶点提供依据。Colorectal cancer is a common human malignant tumour.N6-methyladenine(m6A)modification is considered to be the major internal epigenetic modification in eukaryotic mRNA,the level of m6A modification is dynamic and reversible.It plays a key determining role in the regulation of splicing,translation and degradation of post-transcriptional mRNA.The m6A methyltransferase plays a cancer-promoting or tumor-suppressing role in tumor development,and it has the potential function to become a therapeutic target for malignant tumors.METTL3 is the most important m6A methyltransferase,which several studies have shown to play a role in promoting colorectal cancer through several mechanisms.In this review,we summarized the role and related mechanisms of METTL3 on the initiation and progression of colorectal cancer,for providing a basis for further searching for biomarkers and therapeutic targets.
