宫颈癌患者KDM2A和KDM5A蛋白水平与高危型HPV感染的相关性分析

摘　　要：目的探究宫颈癌患者组蛋白赖氨酸脱甲基化酶2A(KDM2A)和组蛋白赖氨酸去甲基化酶5A(KDM5A)蛋白水平,并分析其水平与高危型人乳头瘤病毒(HR-HPV)感染的相关性。方法选取2021年1月-2024年1月收治的78例宫颈癌患者进行研究,作为宫颈癌组,同期选择宫颈上皮内瘤变(CIN)患者72例作为CIN组,选择76例健康体检者作为对照组。收集受检者一般资料,采用酶联免疫吸附法(ELISA)检测三组血清KDM2A和KDM5A水平,比较CIN组不同分级中血清KDM2A和KDM5A水平,比较CIN组不同分级宫颈癌组不同分期患者的HR-HPV感染情况,分析血清KDM2A和KDM5A水平与宫颈癌患者临床病理关系。结果宫颈癌组血清KDM2A和KDM5A水平高于CIN组及对照组(P<0.05);CIN组不同分级中血清KDM2A和KDM5A水平存在差异(P<0.05);FIGOΙ~Ⅱ期及FIGOⅢ期宫颈癌患者HR-HPV总感染率高于CINΙ级及CINⅡ级,FIGOⅢ期宫颈癌患者HR-HPV总感染率高于CINⅢ级(P<0.05);血清KDM2A和KDM5A水平与宫颈癌患者HPV病毒负荷量、肿瘤分化程度及FIGO分期相关(P<0.05)。结论宫颈癌患者血清KDM2A和KDM5A水平升高,与HR-HPV感染相关。Objective To investigate the protein levels of histone lysine demethylase 2A(KDM2A)and histone lysine demethylase 5A(KDM5A)in cervical cancer patients,and to analyze their correlation with high-risk human papillomavirus(HR-HPV)infection.Methods A total of 78cervical cancer patients admitted from January 2021to January 2024were selected as the cervical cancer group.During the same period,72patients with cervical intraepithelial neoplasia(CIN)were selected as the CIN group,and 76healthy individuals were selected as the control group.The general information of the examinee was collected.Enzyme linked immunosorbent assay(ELISA)was applied to detect serum levels of KDM2Aand KDM5Ain three groups.The serum levels of KDM2Aand KDM5Awere compared among different grades of the CIN group.The HR-HPV infection status of patients with different grades and stages of cervical cancer in the CIN group was compared.The relationship between serum levels of KDM2Aand KDM5Aand clinical pathological characteristics of cervical cancer patients was analyzed.Results The serum levels of KDM2Aand KDM5A in the cervical cancer group were higher than those in the CIN group and control group(P<0.05).There were differences in serum KDM2Aand KDM5Alevels among different grades in the CIN group(P<0.05).The total infection rates of HR-HPV in FIGO stage I-II and FIGO stage III cervical cancer patients were higher than those in CIN stageΙand CIN stage II,the total infection rate of HR-HPV in FIGO stage III cervical cancer patients was higher than that in CIN stage III(P<0.05).The serum levels of KDM2Aand KDM5A were correlated with HPV viral load,tumor differentiation degree,and FIGO staging in cervical cancer patients(P<0.05).Conclusion The serum levels of KDM2Aand KDM5A are elevated in cervical cancer patients,which are associated with HR-HPV infection.
