摘　　要：目的探讨circTRRAP在结直肠癌中的表达水平及临床意义。方法选取首都医科大学附属北京朝阳医院2016年9月至2019年7月收治的67例结直肠癌患者的结直肠癌组织及癌旁正常组织。采用实时定量聚合酶链反应检测circTRRAP在结直肠癌组织及癌旁正常组织中的表达水平,分析其表达水平与结直肠癌患者临床病理特征的关系,利用si RNA干扰circTRRAP的表达水平,通过体外细胞功能实验分析circTRRAP对结直肠癌细胞表型的影响。结果结直肠癌组织中circTRRAP表达量高于癌旁正常组织(P<0.05)。低表达组与高表达组肿瘤浸润深度、淋巴结转移、远处转移、TNM分期比较,差异有统计学意义(P<0.05)。体外实验显示:细胞的增殖能力、迁移及侵袭能力下降(P<0.05)。结论Circ TRRAP在结直肠癌中的表达上调,并与不利的临床病理特征相关,circTRRAP可能发挥促癌作用。Objective To explore the expression level of circTRRAP and its clinical significance in colorectal cancer.Methods Colorectal cancer tissues and adjacent normal tissues of 67 patients with colorectal cancer were selected from September 2016 to July 2019 in Beijing Chao-Yang Hospital,Capital Medical University.Real-time quantitative polymerase chain reaction was used to detect the expression level of circTRRAP in colorectal cancer tissues and adjacent normal tissues.The relationship between the expression level of circTRRAP and the clinicpathological features of colorectal cancer patients was analyzed.The effect of circTRRAP on the phenotype of colorectal cancer cells was analyzed in vitro experiment by interfering or improved the expression level of circTRRAP by siRNA.Results The expression of circTRRAP in colorectal cancer tissues was higher than that in adjacent normal mucosa tissues(P<0.05).There were significant differences in depth of tumor invasion,lymph node metastasis,distant metastasis and TNM staging between low expression group and high expression group(P<0.05).In vitro experiment showed that proliferation,migration,and invasion ability of cells decreased(P<0.05).Conclusion CircTRRAP expression is up-regulated in colorectal cancer.The expression of circTRRAP in colorectal cancer tissues is higher than that in adjacent normal mucosa tissues.circTRRAP may play a role in promoting cancer.
