摘 要:目的:探讨连翘苷(Phil)调节CXC趋化因子配体12(CXCL12)/CXC趋化因子受体4(CXCR4)信号通路对慢性盆腔炎(CPID)大鼠的影响。方法:采用机械损伤联合子宫腔注入混合菌液构建CPID大鼠模型,将造模成功的大鼠分为CPID组、Phil低、中、高剂量组(Phil-L、Phil-M、Phil-H组)和抑制剂组(CXCL12/CXCR4信号通路抑制剂AMD3100),每组12只。另选12只大鼠作为对照组(Sham组)。采用HE染色观察大鼠子宫组织病理学变化并进行病理评分。采用TUNEL染色观察大鼠子宫组织细胞凋亡情况。采用免疫组织化学法检测大鼠子宫组织中剪切的半胱天冬氨酸蛋白酶-1(Cleaved caspase-1)表达。采用ELISA法检测大鼠血清白细胞介素(IL)-6、肿瘤坏死因子(TNF-α)、IL-1β水平。采用Western blot检测大鼠子宫组织Bcl-2相关X蛋白(Bax)、B细胞淋巴瘤-2(Bcl-2)、CXCL12、CXCR4蛋白表达。结果:Sham组大鼠上皮细胞紧密排列,未见明显病理损伤。与Sham组比较,CPID组大鼠子宫组织出现腺体、纤维结缔组织增生,水肿明显,内膜扩张充血,并有大量炎症因子浸润。与CPID组比较,Phil-L组、Phil-M组、Phil-H组、抑制剂组大鼠炎症浸润和纤维结缔组织增生依次减少,且Phil-H组、抑制剂组未见纤维结缔组织增生。与Sham组比较,CPID组大鼠子宫组织病理评分、子宫组织细胞凋亡率、子宫组织Cleaved caspase-1阳性表达率、血清IL-6、IL-1β和TNF-α水平以及子宫组织Bax、CXCL12、CXCR4蛋白表达水平升高,Bcl-2蛋白表达水平降低(均P<0.05)。与CPID组比较,Phil-L、Phil-M、Phil-H组大鼠子宫组织病理评分、子宫组织细胞凋亡率、子宫组织Cleaved caspase-1阳性表达率、血清IL-6、IL-1β和TNF-α水平以及子宫组织Bax、CXCL12、CXCR4蛋白表达水平降低,Bcl-2蛋白表达水平升高(均P<0.05)。Phil-H组上述指标与抑制剂组比较差异无统计学意义(均P>0.05)。结论:Phil能够改善CPID大鼠子宫组织病理损伤,降低细胞�Ob jective:To investigate the effects of phillyrin(Phil)on chronic pelvic inflammatory disease(CPID)rats by regulating CXC chemokine ligand 12(CXCL12)/CXC chemokine receptor 4(CXCR4)signaling pathway.Methods:The CPID rat model was established by mechanical injury combined with uterine cavity injection of mixed bacterial solution.The successful model rats were divided into CPID group,Phil low,medium and high dose groups(Phil-L,Phil-M and Phil-H groups),and inhibitor group(CXCL12/CXCR4 signaling pathway inhibitor AMD3100),with 12 rats in each group.Another 12 rats were selected as the control group(Sham group).HE staining was used to observe the pathological changes of rat uterus and pathological score was performed.TUNEL staining was used to observe the apoptosis of rat uterine tissue.The expression of Cleaved caspase-1 in rat uterine tissue was detected by immunohistochemistry.The serum levels of interleukin(IL)-6,tumor necrosis factor(TNF-α)and IL-1βwere detected by ELISA.Western blot was used to detect the protein expressions of Bcl-2 associated X protein(Bax),B cell lymphoma-2(Bcl-2),CXCL12 and CXCR4 in rat uterine tissue.Results:The epithelial cells in the Sham group were closely arranged,and no obvious pathological damage was observed.Compared with Sham group,the uterine tissue of CPID group showed glands and hyperplasia of fibrous connective tissue,obvious edema,dilation and congestion of endometrium,and infiltration of a large number of inflammatory factors.Compared with the CPID group,the inflammatory infiltration and hyperplasia of fibrous connective tissue in the Phil-L group,Phil-M group,Phil-H group and inhibitor group were reduced in sequence,and no fibrous connective tissue hyperplasia was observed in the Phil-H group and inhibitor group.Compared with Sham group,the pathological score of uterine tissue,apoptosis rate of uterine tissue,positive expression rate of Cleaved caspase-1 in uterine tissue,serum levels of IL-6,IL-1βand TNF-α,and the expression levels of Bax,CXCL12 and CXCR4 proteins in u
