富含脂肪的组织最优冰冻切片条件探索

摘  要:目的探讨在冷冻切片中,不同切片条件对富含脂肪组织切片质量的影响。方法对临床送检的富含脂肪组织,在不同冷冻温度(-30°C、-39°C、-47°C)和切片厚度(10µm、20µm、30µm)条件下进行冷冻切片,并比较其HE染色和快速免疫染色的质量差异。结果在-47°C的低温环境下,切片更加容易完成,且10µm的切片厚薄均匀性、形态完整性及染色对比度均优于其他温度和厚度条件。结论-47°C的冷冻温度和10µm的切片厚度是处理富含脂肪组织冷冻切片的最优条件,可显著提高切片质量。Objective To investigate the slicing quality of adipose-rich tissues under different slicing methods in frozen sections.Methods The quality difference of HE staining and rapid immunostaining results of frozen sections of fatty tissues sent for clinical inspection at different freezing temperatures(-30°C,-39°C,-47°C)and different slicing thicknesses(10μm,20μm,30μm).Results Under the low temperature environment below-47°C,it was easier to obtain thinner and more complete slices.Slices of 10μm thickness obtained at this temperature were more uniformly thin compared to other temperatures and other thicknesses,with better staining contrast.Conclusion Adipose-rich tissues yield more excellent slices of higher quality when sliced at-47°C and a thickness of 10μm.
