蛋白酶高效催化体系构建及其对羊毛角蛋白的降解

摘  要:角蛋白内二硫键结构严重阻碍蛋白酶对其高效降解,为提高酶对角蛋白的降解效率,分别构建了由蛋白酶Savinase 16L与有机膦化合物LKZ-610、β-巯基乙醇、亚硫酸氢钠、L-半胱氨酸组成的4种催化体系。以羊毛鳞片角蛋白为研究对象,采用5,5’-二硫代双(2-硝基苯甲酸)(DTNB)模拟反应、羊毛表面巯基测定及阿尔瓦登反应研究不同二硫键还原剂与二硫键的反应性,通过紫外可见分光光度计、扫描电子显微镜及X射线衍射仪等分析了催化体系对羊毛鳞片角蛋白的降解行为。结果表明:所选取的4种还原剂均可断开二硫键,其中LKZ-610可瞬时打开模拟物DTNB中98.12%的二硫键,其处理后的羊毛表面巯基含量达13.64μmol/g。对比不同催化体系对羊毛鳞片角蛋白的降解效果,优选出由LKZ-610与蛋白酶Savinase 16L组成的高效催化体系,其可在50 min内完全降解羊毛鳞片层,处理后羊毛织物质量损失率达35.81%,结晶指数由20.67%显著降低至4.25%。The disulfide bond structure in keratin severely hinders its efficient degradation by proteases.To improve the efficiency of keratin degradation by proteases,four integrated catalytic systems consisting of the protease Savinase 16L with the disulfide bond reducers LKZ-610,β-mercaptoethanol,sodium bisulfite,and L-cysteine,were constructed respectively.The reactivity of disulfide bond with different reducing agents was studied using 5,5′-dithiobis(2-nitrobenzoic acid)simulation reaction,the determination of sulfhydryl groups on wool surface and Alvaden reaction.The degradation behavior of keratin on wool scales was analyzed by ultraviolet visible spectrophotometer,scanning electron microscope and X-ray diffractometer.The results show that all the four reducing agents could break the disulphide bond,among which LKZ-610 could break 98.12%of the disulphide bond in the simulated DTNB instantaneously,and the content of sulfhydryl group on the surface of the treated wool reached 13.64μmol/g.Comparing the degradation effects of different catalytic systems on keratin of wool scales,a highly efficient catalytic system consisting of LKZ-610 and protease Savinase 16L was selected,which could completely degrade wool scales within 50 min.After treatment,the weight loss rate of wool fabric reached 35.81%,and the crystallization index decreased significantly from 20.67%to 4.25%.
